Summary: A tumor-targeting, pH/redox dual-responsive nanosystem was designed for the co-delivery of chemo-drugs and siRNA. This nanosystem efficiently delivered DOX and siGCN5 into drug-resistant cancer cells, inhibiting tumor growth significantly and downregulating P-gp epigenetically. In an MDR breast tumor model, DOX and siGCN5 loaded HPMSNs effectively inhibited tumor growth, abolished P-gp expression, and minimized DOX's systemic toxicity.

Efforts to maximize the EPR effect in developing smarter nanomedicines are essential for successful clinical applications.
