Identification of MET fusions in solid tumors: A multicenter, large scale study in China

Wenhui Yang et al.

Summary: MET amplification and exon 14 skipping are recognized oncogenic drivers in various cancers, but MET fusions in most cancer types remain poorly defined. Through a large-cohort study using DNA-based NGS, we identified 37 potentially functional MET fusions with intact tyrosine kinase domain (TKD) in 36 samples. Additionally, we discovered 15 novel MET fusions in five cancer types, with the incidence of novel MET fusions accounting for 40.5% (15/37). Our findings highlight the need for further validation of rare MET fusions and emphasize the potential of DNA-based NGS as a clinical test for common MET fusions.

Hereditary Gastric Cancer: Single-Gene or Multigene Panel Testing? A Mono-Institutional Experience

Mariarosaria Calvello et al.

Summary: Gastric cancer has traditionally been overlooked in hereditary cancer research, but the advent of multigene panel testing has shed light on the involvement of additional genes, particularly those related to homologous recombination repair. Our study identified CDH1 and MSH2 as genes associated with early-onset diffuse and later-onset intestinal gastric cancers, respectively. Multigene panel testing was found to be advantageous in patients with unspecific phenotypes, although it posed challenges in result interpretation.

Population-based targeted sequencing of 54 candidate genes identifies PALB2 as a susceptibility gene for high-grade serous ovarian cancer

Honglin Song et al.

Summary: This study aimed to investigate the contribution of rare deleterious germline variants in candidate genes to ovarian cancer susceptibility. The findings suggest a potential association between PALB2 mutations and increased risk of high-grade serous ovarian cancer, with further research needed to confirm the significance of other genes.

Germline variants in DNA repair genes are associated with young-onset head and neck cancer

Sarah Santiloni Cury et al.

Summary: Genetic predisposition to head and neck carcinomas (HNSCC) and the impact of known risk factors on early-onset disease are not well explored. While early-onset HNSCC is rare, its incidence is increasing. Germline alterations in DNA repair genes may contribute to an increased risk for developing HNSCC at an early age, with potential impact on prognosis.

Whole-exome sequencing reveals germline-mutated small cell lung cancer subtype with favorable response to DNA repair-targeted therapies

Summary: This study identified deleterious variants in cancer-predisposition genes in a significant portion of SCLC patients, impacting medical management and family member testing. Germline genotype was associated with family history of cancer, as well as recurrence-free survival after platinum-based chemotherapy. The findings suggest potential for targeted therapies based on inherited predisposition in SCLC patients.

Genomic profiling in non-small-cell lung cancer in young patients. A systematic review

D. Vinal et al.

Summary: Lung cancer in young patients has unique epidemiological, clinicdemographic, and genomic features compared to older patients, with higher female prevalence, more advanced stages, and specific genomic alterations. While still uncommon in young patients, the use of NGS has revealed germline genomic alterations associated with lung cancer.

Germline mutations and age at onset of lung adenocarcinoma

Karen L. Reckamp et al.

Summary: This study found that patients with germline pathogenic variants such as TP53, EGFR, and BRCA2 may experience an earlier onset of lung adenocarcinoma. Future research may need to consider including carriers of high-risk variants in the criteria for lung cancer screening, which could impact precision therapy and reduce lung cancer mortality through earlier stage diagnosis.
