Antibodies to watch in 2023

Summary: In this article, the key events in the development of commercial monoclonal antibody therapeutics in 2022 are discussed, and events that may occur in 2023 are forecasted. By mid-November, 12 antibody therapeutics had received first approvals in the United States or European Union, and an additional 7 had been approved in China or Japan. Globally, at least 24 investigational antibody therapeutics are undergoing regulatory review. The data show that, excluding antibodies for COVID-19, the late-stage commercial clinical pipeline has grown by approximately 20% in the past year.

Acyl carrier protein tag can enhance tobacco etch virus protease stability and promote its covalent immobilisation

Summary: Fusion expression is commonly used to improve the solubility of recombinant proteins, but the fusion tag often needs to be removed to avoid affecting the structure and activity of the target protein. The tobacco etch virus (TEV) protease, known for its stringent sequence recognition, is frequently employed for tag removal. In this study, fusion of TEV with the acyl carrier protein from E. coli fatty acid synthase (ACP) significantly increased the yield and stability of soluble TEV. ACP-TEV maintained high cleavage activity and could be efficiently modified by co-expressed AcpS, allowing immobilization of TEV through the active holo-ACP. The immobilized TEV exhibited high cleavage activity after long-term storage and multiple reuse cycles, making it a promising, cost-effective, and recyclable biocatalyst for biotechnological research and development.

While most clinical trials show minimal immunogenicity, special caution is needed for highly potent sdAbs. Further research is needed to establish humanization thresholds to maximize human sequence content without compromising binding affinity or stability.

Fusion Tag Design Influences Soluble Recombinant Protein Production in Escherichia coli

Summary: Fusion protein technologies are widely used in biopharmaceutical applications, but they may have negative consequences such as reduced product yield and unwanted changes in expression levels. In this study, we evaluated different N-terminal fusion tag combinations to find an ideal design for a generic fusion tag. The CASPON-tag, a combination of a negatively charged peptide tag, affinity tags, and a caspase-2 cleavage site, showed good performance in enhancing soluble expression and could be efficiently removed.

Brolucizumab: A Newly Developed Anti-VEGF Molecule for the Treatment of Neovascular Age-Related Macular Degeneration
