Correlative Extinction and Single Fluorophore Bleaching Microscopy for Ligand Quantification on Gold Nanoparticles

Background-free four-wave mixing microscopy of small gold nanoparticles inside a multi-cellular organ

Summary: The ability to detect small metallic nanoparticles by optical microscopy inside environmentally relevant species may have a wide impact for ecotoxicology studies. We demonstrate four-wave mixing microscopy on individual small gold nanoparticles inside the hepatopancreas of Oniscus Asellus, a terrestrial isopod, which ingests metals found in the soil. Four-wave mixing microscopy holds great potential to investigate the fate of metal nanoparticles inside biological systems.

Correlative light-electron microscopy using small gold nanoparticles as single probes

Summary: In this study, small gold nanoparticles were used as a single probe for correlative light-electron microscopy (CLEM). The nanoparticles were bound to epidermal growth factor protein and precisely located in human cancer cells using light microscopy with four-wave mixing (FWM). The corresponding transmission electron microscopy images showed high accuracy in the correlation mapping. By reducing systematic errors, the correlation accuracy was improved to below 40 nm, and the localization precision was below 10 nm. Polarization-resolved FWM correlated with nanoparticle shapes, promising future applications in multiplexing through shape recognition. The photostability of gold nanoparticles and the applicability of FWM microscopy to living cells make FWM-CLEM a powerful alternative to fluorescence-based methods.

Quantitative morphometric analysis of single gold nanoparticles by optical extinction microscopy: Material permittivity and surface damping effects

Summary: The study analyzed defect-free ultra-uniform gold nanospheres with diameters of 10 nm and 30 nm, finding that the single crystal dataset and the surface damping parameter g approximately equal to 1 were best able to describe the nanoparticle geometries. The results were in good agreement with transmission electron microscopy observations of the same sample batches.
