Novel scaffold platforms for simultaneous induction osteogenesis and angiogenesis in bone tissue engineering: a cutting-edge approach

Biodegradable BBG/PCL composite scaffolds fabricated by selective laser sintering for directed regeneration of critical-sized bone defects

Summary: The repair of critical-sized bone defects (CSBD) is challenging, and personalized 3D porous scaffolding is considered ideal for this purpose. In this study, a customized porous scaffold was designed based on a body-centered cubic unit for rabbit radius. Borate bioactive glass/polycaprolactone (BBG/PCL) composite scaffolds with different BBG contents were fabricated and characterized for various properties. The results showed that the BBG/PCL composite scaffold with 20% BBG content effectively promoted osteoblast proliferation and repaired the rabbit radius defects. The scaffold exhibited sufficient mechanical strength for long-term CSBD repair, indicating its potential as a promising biomaterial.

Osteogenic differentiation of human adipose-derived mesenchymal stem cells in a bisphosphonate-functionalized polycaprolactone/gelatin scaffold

Summary: Recent trends in bone tissue engineering have focused on developing biomimetic constructs with appropriate mechanical and physiochemical properties. In this study, a new biomaterial scaffold based on a bisphosphonate-containing synthetic polymer combined with gelatin was fabricated. The scaffold showed aligned pores, high porosity, and good biodegradability. It exhibited good cytocompatibility with human Adipose-Derived Mesenchymal Stem Cells (hADMSCs) and promoted mineralization and ALP activity. The scaffold also demonstrated high expression of osteogenic genes, suggesting its potential for bone tissue engineering.

PEGylated gold nanoparticles promote osteogenic differentiation in in vitro and in vivo systems

Summary: This study comprehensively evaluated the effects of PEGylated gold nanoparticles of different sizes on osteogenic differentiation, demonstrating excellent biocompatibility and size-dependent promotion of osteogenic effects on various cell types. Further in vivo testing showed that 45-nm GNPs conjugated with PEG-hydrogel significantly stimulated bone regeneration in rabbits with femur defects, indicating promising applications for clinical translation.
