Retrograde signaling in plants: A critical review focusing on the GUN pathway and beyond

Summary: Plastids communicate with the nucleus through retrograde signaling to regulate nuclear gene expression. Plastid biogenesis and responses of mature chloroplasts to environmental changes are controlled through biogenic and operational controls, respectively. The functions of GUN proteins in retrograde signaling and the controversy surrounding GUN1 are discussed in this review, along with recent progress in understanding chloroplast-derived signals in acclimation response and flavonoid biosynthesis.

Summary: This study explores the potential use of NtMAR1hs as a recessive selective marker for DNA-free genome editing using CRISPR/Cas9-sgRNA ribonucleoprotein complex. The knockout of NtMAR1hs in tobacco plants demonstrated their role in sensitivity to aminoglycoside antibiotics, similar to Arabidopsis MAR1. The study also suggests the presence of additional functional homologs in tobacco.

Summary: Iron is an essential cofactor for all living organisms. The membrane transport mechanisms of iron often use ferrous ions, but uncoordinated or deliberated ferrous ions can initiate Fenton reactions. Fe-III citrate complexes are among the most important forms of iron, especially in plants where they can undergo photoreduction. The photoreaction of ferric species in leaves may have biological relevance in iron metabolism, although the specific relevance is not well understood.

Qualitative and quantitative evaluation of thylakoid complexes separated by Blue Native PAGE

Summary: In this study, the Blue/Clear-Native polyacrylamide gel electrophoresis (BN PAGE) method was improved by using a detergent mixture of 1% (w/V) n-dodecyl-beta-d-maltoside plus 1% (w/V) digitonin for solubilisation and 4.3-8% gel gradients for separation. Several large photosystem (PS) I containing bands were detected, and it was found that these bands were PSI-NADH dehydrogenase-like megacomplexes in maize bundle sheath thylakoids and PSI complexes with different light-harvesting complex (LHC) complements (PSI-LHCII, PSI-LHCII*) in mesophyll thylakoids of lincomycin treated maize. Proposed methods for quantitative determination and comparison of the complexes in different samples were also provided.

The Cluster Transfer Function of AtNEET Supports the Ferredoxin-Thioredoxin Network of Plant Cells

Summary: NEET proteins are conserved 2Fe-2S proteins that regulate iron and reactive oxygen species levels in plant and mammalian cells. Disrupting AtNEET function causes oxidative stress, chloroplast iron overload, activation of iron-deficiency responses, and cell death. This study reveals new roles for AtNEET in supporting the ferredoxin-thioredoxin network, iron mobilization, and cytosolic 2Fe-2S cluster assembly in plants.

Aggregation-related quenching of LHCII fluorescence in liposomes revealed by single-molecule spectroscopy

Summary: This study investigated the fluorescence properties of liposome-reconstituted major light-harvesting complexes of plants (LHCII) using fluorescent dye labelling and single-molecule microscopy techniques. Results showed that the fluorescence lifetimes in bulk and single liposome measurements were correlated, with shorter lifetimes observed in liposome-free LHCII compared to reconstituted LHCII. The dependence of fluorescence lifetime on protein density in liposome-reconstituted LHCII resembled concentration quenching, while the impact of liposome size on fluorescence lifetime was not significant. It was demonstrated that fluorescence quenching in reconstituted membranes could be induced by LHCII-LHCII interactions, similar to photoprotective non-photochemical quenching in vivo.
