Killer to cure: Expression and production costs calculation of tobacco plant-made cancer-immune checkpoint inhibitors

Laura A. Ridgley et al.

Summary: Immune checkpoint inhibitors (ICIs) have shown significant clinical success, but their response rates are limited and they are expensive. This study successfully expressed three key ICIs transiently in Nicotiana benthamiana and Nicotiana tabacum plants. The ICIs were characterized in terms of protein levels, target cell binding, receptor binding, and recovery during purification. The study suggests that plants can be potential alternative platforms for producing affordable and accessible ICIs.

A plant-produced SARS-CoV-2 spike protein elicits heterologous immunity in hamsters

Emmanuel Margolin et al.

Summary: By applying an integrated host and glyco-engineering approach, a model antigen of the emerging virus SARS-CoV-2 was successfully produced in Nicotiana benthamiana. The protein produced in plants had a similar structure but differed in glycosylation compared to the protein produced in mammalian cells. Animals immunized with the plant-derived antigen showed reduced viral loads, lower lung pathology, and protection from weight loss after challenge, although the protection was not as potent as that induced by the mammalian cell-culture-derived antigen.

Comparative analysis of plant transient expression vectors for targeted N-glycosylation

Lukas Eidenberger et al.

Summary: This study investigated the capability of three commonly used transient expression vectors to express a multi-component protein with controlled and modified N-glycosylation. The results showed that all three expression vectors produced similar quantities of targeted N-glycosylation, demonstrating the universal application of plant-based glycoengineering.

Harnessing the Potential of Plant Expression System towards the Production of Vaccines for the Prevention of Human Papillomavirus and Cervical Cancer

Balamurugan Shanmugaraj et al.

Summary: Cervical cancer is a common gynecological malignant tumor, especially in developing countries, and it remains a major health problem. Despite research efforts, cervical cancer remains the leading cause of cancer death. Human papillomavirus (HPV) is the most important risk factor for cervical cancer. Early detection and immunotherapies show promise in improving survival rates, but high cost limits equal access to HPV vaccines. Developed countries have implemented screening programs and disease control systems, but low-income and middle-income countries see increasing incidence and limited access to affordable therapeutic options. Plant expression platforms offer a possible solution for producing affordable HPV vaccines.

Increased in vitro neutralizing activity of SARS-CoV-2 IgA1 dimers compared to monomers and IgG

Lin Sun et al.

Summary: In this study, two neutralizing monoclonal antibodies against SARS-CoV-2 were expressed in glycoengineered Nicotiana benthamiana plants in three formats. The IgA1 dimers showed a significantly higher neutralization potency than monomers, indicating the positive impact of IgA structural features and multivalency on neutralization potency. Additionally, the study highlighted the versatile use of plants for rapid expression of complex human proteins.
