TRIM3 attenuates cytokine storm caused by Dabie bandavirus via promoting Toll-like receptor 3 degradation

TRIM7 inhibits encephalomyocarditis virus replication by activating interferon-? signaling pathway

Summary: Tripartite motif-containing protein 7 (TRIM7) plays an important role in innate immune responses against viral infection, particularly Encephalo-myocarditis virus (EMCV) infection. TRIM7 inhibits EMCV replication through the type I interferon (IFN) signaling pathway. Overexpression of TRIM7 suppresses EMCV replication and enhances IFN-I3 promoter activity in HEK293T cells, while knockdown of endogenous TRIM7 promotes EMCV infection and impairs IFN-I3 promoter activity. TRIM7 regulates IFN-I3 signaling pathway mediated by retinoic acid-inducible gene I (RIG-I)/ melanoma differentiation-associated gene 5 (MDA5)/ mitochondrial antiviral-signaling protein (MAVS). TRIM7 interacts with MAVS and co-localizes in HEK293T cells. These findings demonstrate that TRIM7 plays a positive role in the IFN-I3 signaling pathway during EMCV infection and acts as a suppressor of EMCV replication. The results suggest that TRIM7 could be a potential target for the development of anti-EMCV inhibitors.

Summary: Chinese rhesus macaques infected with SFTSV show mild clinical symptoms similar to human SFTS, including slight fever, thrombocytopenia, leukocytopenia, and increased AST and CK levels in the blood. During infection, the virus persists in lymphoid tissues and bone marrow, and there are changes in the immune cell population. This study provides valuable insights into the pathogenesis of SFTSV infection.

Summary: The activation of the NLRP3 inflammasome and induction of pyroptosis, leading to increased secretion of IL-1 beta/IL-18, play important roles in viral pathogenesis and host protection during severe fever with thrombocytopenia syndrome virus (SFTSV) infection. Pyroptosis, as part of innate immunity, might be essential in proinflammatory responses and pathogenicity in humans infected with this novel phlebovirus.

TRIM3 facilitates estrogen signaling and modulates breast cancer cell progression

Summary: In this study, the researchers found that TRIM3, an E3 ligase, promotes ER alpha signaling activity and breast cancer progression. Depletion of TRIM3 inhibits breast cancer cell proliferation and migration, and unbiased RNA sequencing data showed that TRIM3 is crucial for estrogen signaling activity on a genome-wide scale. The researchers also demonstrated that TRIM3 associates with ER alpha and promotes its stability by inducing a specific ubiquitin modification. These findings provide a novel posttranslational mechanism in estrogen signaling and suggest that modulation of TRIM3 expression or function could be a promising approach for breast cancer treatment.
