Microbial nanowires: type IV pili or cytochrome filaments?

Summary: A new field of study on long-range electron transport by extracellular filaments in anaerobic bacteria has emerged, with Geobacter sulfurreducens being the model system. This area of research is of interest due to the potential applications in bioremediation, energy generation, and bio-based nanotechnology. Recent atomic structures of the T4P from G. sulfurreducens obtained by cryo-electron microscopy (cryo-EM) have challenged previous models of electron conduction, revealing that conductive filaments are actually polymerized cytochromes or extracellular DNA. It is believed that similar polymerized cytochromes may be found in other anaerobic organisms.

Structure of Geobacter cytochrome OmcZ identifies mechanism of nanowire assembly and conductivity

Summary: By using cryogenic electron microscopy structure, we revealed the linear and closely stacked haems of OmcZ nanowires, which may account for its electron conductivity. The surface-exposed haems and charge interactions explained the binding interactions between OmcZ nanowires and diverse extracellular electron acceptors, as well as the rearrangement of nanowire network in different biochemical environments. In vitro studies demonstrated how G. sulfurreducens employed a serine protease to control the assembly of OmcZ monomers into nanowires. We found that OmcZ and serine protease are widespread in environmentally important bacteria and archaea, establishing the prevalence of nanowire biogenesis across diverse species and environments.

Summary: The model electroactive bacterium Geobacter sulfurreducens can acquire electrons directly from solid donors and use C4 dicarboxylates as a carbon source. Mutations in the PilR regulator and the ability to utilize solid electron donors contribute to the competitive advantage and widespread distribution of G. sulfurreducens in anaerobic ecosystems.

The roles of DmsEFAB and MtrCAB in extracellular reduction of iodate by Shewanella oneidensis MR-1 with lactate as the sole electron donor

Summary: The roles of gene clusters dmsEFAB, mtrCAB, mtrDEF, and so4360-4357 in the extracellular reduction of iodate with lactate as an electron donor were investigated in Shewanella oneidensis MR-1. Deletions of dmsEFAB and/or mtrCAB gene clusters reduced the ability of bacteria to reduce iodate. DmsEFAB and MtrCAB worked together to reduce iodate, with DmsEFAB playing a more dominant role. MtrCAB was involved in detoxifying hydrogen peroxide, and the reaction intermediate hypoiodous acid was found to inhibit microbial iodate reduction. SO4360-4357 and MtrDEF were not involved in iodate reduction.

Cryo-EM structure of an extracellular Geobacter OmcE cytochrome filament reveals tetrahaem packing

Summary: Two electrically conductive appendages are produced by the bacterium Geobacter sulfurreducens. One is a conductive fiber containing polymerized c-type cytochrome OmcS subunits, and the other is a thinner appendage comprised of cytochrome OmcE subunits. Both filaments share a conserved haem packing arrangement in which the haems are coordinated by histidines in adjacent subunits, despite having no overall sequence or structural similarities.

Structure of Geobacter OmcZ filaments suggests extracellular cytochrome polymers evolved independently multiple times

Summary: Early genetic and structural observations suggested that metal-reducing organisms like Geobacter had extracellular conductive filaments composed of type IV pili. However, recent research has shown that bacterial c-type cytochromes can also form extracellular filaments for long-range electron transport. Cryo-electron microscopy was used to determine the structure of a third extracellular filament, formed from Geobacter sulfurreducens octaheme cytochrome, OmcZ. The structure of OmcZ differs from that of OmcS and OmcE, indicating that conductive cytochrome polymers likely arose independently from different ancestral multiheme proteins.

Structure of Geobacter pili reveals secretory rather than nanowire behaviour

Summary: Geobacter species transfer electrons extracellularly through microbial nanowires known as surface appendages. Recent studies have revealed that the nanowires consist of cytochromes OmcS and OmcZ, rather than the previously believed pili composed of PilA-N protein. The use of PilA-C in assembling heterodimeric pili resembles the structure and function of type 2 secretion pseudopili, indicating a potential widespread effect on diverse electron-transferring microorganisms.
