Versatility of bacterial outer membrane vesicles in regulating intestinal homeostasis

Xinyue Wang et al.

Summary: Outer membrane vesicles (OMVs) released by Akkermansia muciniphila can restore gut microbiota balance, stimulate mucosal immunoglobulin A response, and maintain intestinal barrier integrity by entering intestinal epithelial cells. Transplanting gut microbiota-derived OMVs can alleviate colitis and enhance anti-PD-1 therapy against colorectal cancer. This study highlights the important role of gut microbiota-derived OMVs in intestinal ecology and provides a new target for disease intervention and treatment.

Outer-Membrane Vesicles of Fusobacterium necrophorum: A Proteomic, Lipidomic, and Functional Characterization

Prabha Bista et al.

Summary: OMVs from Fusobacterium necrophorum subspecies necrophorum, an opportunistic necrotic infection-causing pathogen, were purified and characterized. The analysis revealed the presence of various outer-membrane proteins and toxins in OMVs, as well as their high cytotoxicity. OMVs mainly consisted of phospholipids, sphingolipids, and acetylcarnitine. Further studies are needed to evaluate OMVs' ability to induce immune responses and their potential as vaccines.

Inflammatory modulation and outer membrane vesicles (OMV) production associated to Bacteroides fragilis response to subinhibitory concentrations of metronidazole during experimental infection

Michele Cristine Ribeiro de Freitas et al.

Summary: This study reproduced an experimental infection of Bacteroides fragilis using a tissue cage model to evaluate the subinhibitory concentration (SIC) of metronidazole in OMV production patterns and immunological and histological characteristics. The results showed that in the experimental group, the total counts of leukocytes and neutrophils were higher, and there was an exacerbated inflammatory infiltrate. Additionally, there was a massive production and release of OMV. These results confirm the potential involvement of OMV in the pathogenicity of B. fragilis.

Translocated Legionella pneumophila small RNAs mimic eukaryotic microRNAs targeting the host immune response

Tobias Sahr et al.

Summary: This study reveals that Legionella pneumophila mimics eukaryotic microRNA to modulate the host response to infection. This miRNA-like regulation is a feature of host-pathogen communication and may represent a general mechanism used by bacteria that interact with eukaryotic hosts.

The Legionella pneumophila Dot/Icm type IV secretion system and its effectors

Daniel C. Lockwood et al.

Summary: To gain an advantage in interacting with eukaryotic hosts, many bacterial pathogens use protein secretion systems to release virulence factors. Legionella pneumophila's Dot/Icm T4BSS is an example of a complex and sophisticated secretion system that allows the bacteria to evade degradation and replicate within host cells. Recent advancements in imaging large protein complexes have provided new insights into the structure and transport mechanism of the T4BSS, as well as the functions of L. pneumophila effectors.

RNA Sequencing Unveils Very Small RNAs With Potential Regulatory Functions in Bacteria

Idrissa Diallo et al.

Summary: This study provides a detailed analysis of very small RNAs (vsRNAs) in Escherichia coli, E. coli-derived outer membrane vesicles (OMVs), and five other bacterial strains. The results show that 8-15 nt vsRNAs are more abundant than 16-30 nt vsRNAs. tRNA-derived fragments (tRFs), particularly Ile-tRF and Ala-tRF, were found to be major vsRNA types and selectively loaded in OMVs.

Zinc metalloprotease ProA of Legionella pneumophila increases alveolar septal thickness in human lung tissue explants by collagen IV degradation

Lina Scheithauer et al.

Summary: ProA, a secreted zinc metalloprotease of Legionella pneumophila, plays a critical role in promoting infection and tissue damage in human lung tissue, mainly through extracellular activity. It contributes to pathogen dissemination and serum resistance by targeting a wide range of substrates, with distinctive features in substrate binding pocket and cofactor repertoire compared to homologues.

Staphylococcus aureus membrane vesicles contain immunostimulatory DNA, RNA and peptidoglycan that activate innate immune receptors and induce autophagy

Natalie J. Bitto et al.

Summary: Gram-positive bacteria produce membrane vesicles (MVs) containing microbe-associated molecular patterns (MAMPs) such as proteins, nucleic acids, and peptidoglycan, which can activate pattern recognition receptors (PRRs) in host epithelial cells and promote cytokine and chemokine release. Additionally, MVs can be degraded intracellularly through autophagy, enhancing our understanding of their immunostimulatory roles and fate within the host.

Microbial vesicle-mediated communication: convergence to understand interactions within and between domains of life

Hannah M. McMillan et al.

Summary: Extracellular vesicles (EVs) produced by all cells play important roles in interactions between microbial EVs, plants, and nutrient cycling in the environment. Studies have identified molecules within EVs and their surfaces that contribute to these functions, while research on engineered nanomaterials helps understand the impact of surface features on transport and function. Interdisciplinary approaches involving biology, soil geochemistry, and colloid science are essential for fully understanding EV-mediated interkingdom interactions.
