Origin, specification and differentiation of a rare supporting-like lineage in the developing mouse gonad

Chloe Mayere et al.

Summary: This study investigated an uncharacterized population of supporting-like cells (SLCs) in the developing mouse gonads, focusing on their origin, specification, and subsequent sex-specific differentiation. The SLC lineage, closely related to the coelomic epithelium, was specified as early as E10.5, making it the first somatic lineage in the bipotential gonad. SLCs progressively acquired a more Sertoli- or pregranulosa-like identity and contributed to the formation of the rete testis and rete ovarii. WNT4 was found to be a crucial regulator of the SLC lineage and necessary for normal development of the rete testis.

Summary: The study generated a comprehensive map of human and mouse gonadal differentiation using single-cell and spatial transcriptomics, chromatin accessibility assays, and fluorescent microscopy. It identified human-specific regulatory programs and resolved the cellular and molecular events in gonadal development, providing guidance for in vitro gonadogenesis.

Identification of regulatory elements required for Stra8 expression in fetal ovarian germ cells of the mouse

Chun-Wei Feng et al.

Summary: This study demonstrates the crucial role of retinoic acid responsive elements (RAREs) in initiating Stra8 expression in the fetal mouse ovary, along with the enhancement of Stra8 expression by the transcription factor DMRT1 in the presence of retinoic acid. The specific anterior-to-posterior pattern of Stra8 initiation in germ cells is faithfully recapitulated by the 2.9 kb Stra8 promoter, highlighting the importance of RAREs in regulating germ cell meiosis.

Primary sex determination in birds depends on DMRT1 dosage, but gonadal sex does not determine adult secondary sex characteristics

Jason Ioannidis et al.

Summary: In birds, sex determination is thought to depend on a sex chromosome gene dosage mechanism, with the gene DMRT1 playing a key role. Experiments showed that ZZ chickens lacking DMRT1 developed ovaries instead of testes, while blocking estrogen synthesis in ZW embryos led to testis development, highlighting the interconnected roles of DMRT1 and estrogen in avian sex determination.

The conserved sex regulator DMRT1 recruits SOX9 in sexual cell fate reprogramming

Robin E. Lindeman et al.

Summary: The study highlighted the importance of the sex regulator DMRT1 and the maintenance of cell fate in sexual development, showing that DMRT1 can reprogram female granulosa cells into Sertoli-like cells in vivo and in culture. These findings reveal the key roles of chromatin state, gene expression, and different growth factors in the process of sexual cell differentiation.

Arrest of WNT/β-catenin signaling enables the transition from pluripotent to differentiated germ cells in mouse ovaries

Morgane Le Rolle et al.

Summary: This study shows that germ cell-intrinsic beta-catenin activity in mouse embryonic ovaries is essential for maintaining pluripotency and its repression is crucial for differentiation and meiosis entry. The interaction of beta-catenin with the pluripotent-associated factor POU5F1 in the nucleus is associated with germ cell pluripotency, and its exit from the nucleus correlates with germ cell differentiation. These findings highlight the role of beta-catenin as a central gatekeeper in ovarian differentiation and gametogenesis.
