Molecular mechanisms of environmental exposures and human disease

Summary: A substantial proportion of disease risk for common complex disorders is attributable to environmental exposures and pollutants. Recent research has shown the impact of environmental pollutants on the human genome and epigenome through epigenetic modifications. Furthermore, other molecular processes, such as the production of extracellular vesicles, epitranscriptomic changes, and mitochondrial changes, have also been identified as mechanisms through which the environment can exert its effects.

DNA methylation as a potential mediator of the association between indoor air pollution and neurodevelopmental delay in a South African birth cohort

Summary: This study found that exposure to indoor air pollution during pregnancy is associated with neurodevelopmental delay in infants, and DNA methylation might explain this link. Through high-dimensional mediation analysis methods, differentially methylated CpG sites and genes that mediate the association between indoor air pollution and neurodevelopmental delay were identified. The findings suggest that DNA methylation may mediate the association between prenatal PM10 exposure and cognitive neurodevelopment.

Cross-tissue correlations of genome-wide DNA methylation in Japanese live human brain and blood, saliva, and buccal epithelial tissues

However, assessing DNA methylation in brain tissue is challenging due to tissue specificity. These findings are valuable for interpreting epigenetic research in Asian populations.

The link between epigenetic remodeling and the glioma TME provides insights for the development of epigenetic-targeting therapies to improve antitumor immune response.

Understanding epigenetics can be applied to diagnosing and treating metabolic diseases, including the use of epigenetic biomarkers, drugs, and editing.
