Developing a Chromochloris zofingiensis Mutant for Enhanced Production of Lutein under CO2 Aeration

Summary: In this study, a Chromochloris zofingiensis mutant (Cz-pkg) that stays green upon glucose treatment was generated and characterized. The mutant accumulated lutein up to 31.93 +/- 1.91 mg L-1, which is much higher than the common commercial source. Transcriptomic analysis revealed that the dysfunctional cGMP-dependent protein kinase (PKG) in Cz-pkg may regulate the metabolisms of photosynthetic pigments.

By optimizing culture conditions and adding gibberellic Acid-3 (GA3) as well as increasing the C/N ratio and NaCl concentration, the researchers achieved high biomass and astaxanthin yield, surpassing the previous records.

Rutin Stimulates the Green Alga Chromochloris zofingiensis for Improved Biomass and Astaxanthin Production

Summary: Rutin treatment was found to enhance biomass and astaxanthin accumulation in Chromochloris zofingiensis, resulting in increased astaxanthin production. Rutin-treated C. zofingiensis showed higher levels of chlorophylls, proteins, and lipids and lower carbohydrate levels. Rutin promoted intracellular abscisic acid (ABA) levels, which further increased biomass concentration and astaxanthin accumulation.

Co-production of lutein, zeaxanthin, and 13-carotene by utilization of a mutant of the green alga Chromochloris zofingiensis

Summary: In this study, a mutant of Chromochloris zofingiensis (CZ-LZM3) was obtained, which could achieve ultrahigh cell density and simultaneously accumulate lutein, zeaxanthin, and 13-carotene under heterotrophic conditions. The insertional mutation in the beta-carotene ketolase encoding gene was identified as the cause of astaxanthin biosynthesis blockage in CZ-LZM3. By utilizing a two-stage heterotrophic cultivation strategy, this mutant could produce high levels of lutein, zeaxanthin, and 13-carotene.

With the development of genetic tools, C. zofingiensis has become an emerging model for studying lipid metabolism and carotenogenesis, and strategies for trait improvements in triacylglycerol and astaxanthin synthesis have been discussed.
