tRNA-derived RNAs: Biogenesis and roles in translational control

Identification of RNA helicases with unwinding activity on angiogenin-processed tRNAs

Summary: Stress-induced tRNA fragmentation, catalyzed by endonucleolytic activities, produces tRNA-derived small RNAs (tsRNAs) that play important roles in cell signaling, survival, and gene expression regulation. However, the process by which specific endonuclease-targeted tRNAs are processed into individual and potentially biologically active tsRNAs remains poorly understood. In this study, we identified proteins associated with stress-induced tsRNAs and discovered ATP-dependent RNA helicases that can separate tRNAs with endonuclease-mediated nicks. These findings provide insights into the cellular pathways involved in producing individual tsRNAs and their physiological impact.

Nicked tRNAs are stable reservoirs of tRNA halves in cells and biofluids

Summary: This study reveals the stability and function of nonvesicular extracellular RNAs (nv-exRNAs), particularly tRNA-derived small RNAs (tDRs), in human biofluids. It shows that traditional molecular biology methods can result in the denaturation of nicked tRNAs, leading to the production of artifactual tDRs. Enzymatic repair and chromatographic separation were used to regenerate full-length tRNAs and separate nicked tRNAs from tDRs, respectively.

In Salmonella enterica serovar Typhimurium, nicked tRNAs ending in 2',3' cyclic phosphate are ligands for the RtcR transcriptional activator and contribute to RNA repair. Similarly, in human cells, tRNA fragments can also remain as nicked tRNAs after cleavage. This research provides insights into the function of tRNA fragments in both bacteria and eukaryotes.

Pseudouridine-modified tRNA fragments repress aberrant protein synthesis and predict leukaemic progression in myelodysplastic syndrome

Summary: This study demonstrates that pseudouridylation of a stem cell-enriched tRF subtype, mini tRFs containing a 5' terminal oligoguanine (mTOG), selectively inhibits aberrant protein synthesis programs, thereby promoting engraftment and differentiation of hematopoietic stem and progenitor cells in MDS patients. The mechanism involves hindering the recruitment of translational co-activator PAIP1 and repressing translation of transcripts sharing pyrimidine-enriched sequences at the 5' UTR, including TOP sequences frequently altered in cancer. Dysregulation of mTOG leads to increased translation of 5' PES mRNA in malignant MDS-HSPCs, which is associated with leukaemic transformation and reduced patient survival.

RTCB Complex Regulates Stress-Induced tRNA Cleavage
