The effect of pore size and layout on mechanical and biological properties of 3D-printed bone scaffolds with gradient porosity

Mahsa Karimi et al.

Summary: The design of porous gradient scaffolds for bone tissue engineering is a new approach that imitates bone tissue to enhance cellular responses. Additive manufacturing, such as fused filament fabrication, allows for precise control of pore size. The scaffolds' properties were evaluated, and it was found that higher gradient porosity and larger pore size led to better biological results but lower mechanical strength. Adding hydroxyapatite increased mechanical strength and enhanced cellular response.

An in silico model predicts the impact of scaffold design in large bone defect regeneration

Camille Perier-Metz et al.

Summary: This article uses computer modeling to investigate the mechano-biological principles behind scaffold-guided bone regeneration and the impact of scaffold design on the regeneration process. The study identifies two key factors in scaffold-guided regeneration: the scaffold surface guiding cellular migration and tissue formation processes, and the stimulation of progenitor cell activity by the scaffold material composition. Additionally, a lower scaffold surface-area-to-volume ratio is found to be beneficial for bone regeneration, and a reduced scaffold Young's modulus also favors bone formation.

Characterization and in vitro assessment of three-dimensional extrusion Mg-Sr codoped SiO2-complexed porous microhydroxyapatite whisker scaffolds for biomedical engineering

Chengyong Li et al.

Summary: A microhydroxyapatite whisker scaffold doped with essential trace elements Mg2+ and Sr2+ was developed, showing improved mechanical properties and osteogenic activity. The scaffold had sustained release of osteogenic elements and promoted cell proliferation and osteogenic differentiation. These composite porous scaffolds have the potential to serve as effective bone repair materials.

Regenerative Approaches for the Treatment of Large Bone Defects

Alexander Stahl et al.

Summary: Engineered materials are increasingly used as substitutes for autologous bone grafts in orthopedic practice, but their regenerative efficacy is still limited. Large bone defects greater than 4-5 cm require more complex surgical interventions for repair. Current focus is on improving tissue engineering and vascularization strategies for large bone defect repair.

Large-pore-size Ti6Al4V scaffolds with different pore structures for vascularized bone regeneration

Chao Wang et al.

Summary: The study explored the optimal pore structure and size of Ti6Al4V porous scaffold for repairing large-area bone defects and promoting vascularization in the early stage of osteogenesis. The research found that as pore size increased, cell proliferation and viability also increased, with irregular pore sizes supporting stronger cell vascularization ability compared to regular pore sizes.

Microstructure, shape memory properties, and in vitro biocompatibility of porous NiTi scaffolds fabricated via selective laser melting

H. Z. Lu et al.

Summary: In this study, three porous NiTi scaffolds with different pore sizes but the same porosity were fabricated using selective laser melting (SLM). The scaffolds were systematically studied for their microstructure, mechanical properties, shape memory properties, and in vitro biocompatibility. The results showed that the elastic modulus of the scaffolds was similar to human bones, and the compressive strength and recovery rate were better than most reported porous NiTi alloys.

Camille Perier-Metz et al.

Summary: 3D printed scaffolds are a promising treatment option for large bone defects, with recent design strategies focusing on optimizing geometry and mechanical stimulation for bone formation. However, providing optimal mechanical conditions immediately after surgery may not always lead to optimal long-term bone regeneration outcomes.
