Improving solubility of poorly water-soluble drugs by protein-based strategy: A review

Summary: Poorly water-soluble drugs pose a significant challenge in pharmaceutical development due to their low solubility, which can result in reduced bioavailability and therapeutic effectiveness. Efforts have been made to improve the solubility of these drugs for better pharmacokinetics and pharmacodynamics. Protein-based strategies, which offer natural advantages such as versatile interactions with hydrophobic drugs, biocompatibility, biodegradation, and protein metabolism, have gained special interest. This review summarizes the advancements in using proteins, including plant proteins, mammalian proteins, and recombinant proteins, to enhance the water solubility of poorly water-soluble drugs.

Characterization of excipients to improve pharmaceutical properties of sirolimus in the supercritical anti-solvent fluidized process

Summary: In this study, the impact of excipients on the pharmaceutical properties of sirolimus in the SAS-FB process was evaluated. Co-precipitation of excipients and sirolimus was found to affect the specific surface area of the carrier and drug yield. Among the tested excipients, the formulation containing polyvinylpyrrolidone K30 showed the highest drug yield and significantly improved oral bioavailability compared to the Rapamune tablet. Characterization of the processed API was conducted to understand the mechanism by which excipients contributed to drug dissolution properties.

CO2 utilization as a supercritical solvent and supercritical antisolvent in production of sertraline hydrochloride nanoparticles

Polymorphism characterization of segesterone acetate: A comprehensive study using XRPD, FT-IR and Raman spectroscopy

The study also proposed a method to quantify the ratio of polymorphs I and II, which could be used as a routine analysis step in the manufacturing process of SA.

Solubility and thermodynamic analysis of famotidine polymorphs in pure solvents
