Virally encoded interleukin-6 facilitates KSHV replication in monocytes and induction of dysfunctional macrophages

Article Macrophages drive KSHV B cell latency

Summary: Kaposi's sarcoma herpesvirus (KSHV) establishes lifelong infection in B cells, but in vitro B cell infection is inefficient and cells rapidly die. This study reveals that KSHV infection of monocytes or M-CSF-differentiated macrophages is highly efficient and induces the production of B cell chemoattractants and activating factors. The presence of these mononuclear phagocytes stimulates B cell survival, proliferation, and differentiation, as well as infected plasma cell differentiation and viral latency. These findings suggest a role for mononuclear phagocytes in KSHV B cell latency and possibly contribute to the geographic disparity of KSHV.
