Summary: Protein kinases play important roles in the adaptation of Candida albicans to iron limitation. The protein kinase Ire1 is crucial for the growth of C. albicans under iron-limiting conditions, as it is required for the localization of the high-affinity iron permease Ftr1 to the cell membrane. Ire1 functions independently of endoplasmic reticulum stress and is essential for the virulence of C. albicans.

Two distinct lipid transporters together regulate invasive filamentous growth in the human fungal pathogen Candida albicans

Summary: Flippases play a critical role in lipid transport and asymmetric distribution. In this study, we found that the mutation of a flippase in Candida albicans led to altered distribution of phospholipids and sterols. Other lipid transporters were not necessary for filamentous growth, but the deletion of Osh4 could bypass the requirement for the flippase in invasive filamentous growth. Furthermore, the results suggested that Drs2 and Sac1 function in parallel pathways.

Summary: Candida albicans is a common cause of human mucosal yeast infections, and antifungal medications targeting the pathogen cell wall or plasma membrane have been effective. This study shows that the transport of glucosylceramide (GIcCer) and/or phosphatidylcholine (PC) by plasma membrane P4-ATPases is important for the pathogenicity of Candida albicans. The mutant strains lacking Dnf2 protein exhibit defects in yeast-to-hypha growth transition, filamentous growth, and virulence in systemically infected mice.

This mutation enabled Kis1 to bind to the catalytic alpha-subunit Snf1 in the absence of Snf4, restoring SNF1-dependent gene expression and providing new insights into the regulation of repressors Mig1 and Mig2 in the metabolic adaptation of C. albicans.
