Real-Time Imaging of the Mechanobactericidal Action of Colloidal Nanomaterials and Nanostructured Topographies

Felipe Viela et al.

Summary: Mechanobactericidal nanomaterials are promising antimicrobial strategies that rely on mechanical interactions with bacteria and overcome antibiotic resistance. However, the detailed mechanisms behind the killing effects are not well understood. This study investigates the real-time response of bacteria to weak mechanical interactions with nanostructured topographies and nanodarts, shedding light on the complex mechanisms of mechanically induced bacterial death.

Programmed Death of Injured Pseudomonas aeruginosa on Mechano-Bactericidal Surfaces

Shuo Zhao et al.

Summary: This study reveals that mechanical injury alone is not enough to immediately kill bacteria, instead causing a type of apoptosis-like death. Removal of mechanical stress leads to poststress apoptosis-like death in damaged cells due to self-accumulated reactive oxygen species in a nonstressed environment.

Min Oscillations as Real-time Reporter of Sublethal Effects in Photodynamic Treatment of Bacteria

Ingrid Ortega et al.

Summary: This study investigates the effect of photodynamic treatment on the oscillations of MinD, a cell division regulator in Escherichia coli, using real-time single-cell fluorescence imaging. The results show that the concentration of methylene blue, a photosensitizer, disrupts the oscillation pattern of MinD. In contrast to antibiotics, photodynamic treatment abruptly interrupts the oscillation, reflecting different physiological mechanisms leading to bacterial death.

A guide for membrane potential measurements in Gram-negative bacteria using voltage-sensitive dyes

Jessica A. Buttress et al.

Summary: This paper provides guidance on reliably monitoring membrane potential and its changes in Gram-negative bacteria using the voltage-sensitive dye 3,3'-dipropylthiadicarbocyanine iodide [DiSC 3(5)]. The confounding effects caused by the presence of the outer membrane or measurements performed in buffers are also discussed.

Self-organized protein patterns: The MinCDE and ParABS systems

Adrian Merino-Salomon et al.

Summary: Self-organized protein patterns play a key role in biological decision-making processes, identifying cell division sites, establishing cell polarity, and ensuring DNA segregation. Despite their different functions, the MinCDE and ParABS systems share similar molecular components and mechanisms to achieve biological robustness.

Colistin kills bacteria by targeting lipopolysaccharide in the cytoplasmic membrane

Akshay Sabnis et al.

Summary: Colistin is a last-resort antibiotic with poor efficacy due to growing resistance. Recent research has shown that colistin exerts bactericidal activity by targeting lipopolysaccharide in the cytoplasmic membrane of bacteria. These findings offer insight into novel approaches for enhancing therapeutic outcomes.
