Cryoprotection in Human Mesenchymal Stromal/Stem Cells: Synergistic Impact of Urea and Glucose

Markus Kardorff et al.

Summary: This study aims to develop safe and well tolerated formulations for the (cryo-) preservation of cell therapy products. The cryoprotective capabilities of urea and glucose were evaluated through measurements of cell metabolic activity. Freezing human mesenchymal stromal/stem cells and human dermal fibroblasts at -65°C with equimolar ratios of urea and glucose resulted in comparable viabilities to established dimethyl sulfoxide. Pre-incubation of human mesenchymal stromal/stem cells in trehalose and addition of mannitol and sucrose further enhanced cell viability after freeze-thaw stress. Other cell types assessed could not be satisfactorily preserved with urea and glucose, highlighting the need for tailored formulations.

Evaluating the Use of TiO2 Nanoparticles for Toxicity Testing in Pulmonary A549 Cells

Jana Bacova et al.

Summary: The study optimized the experimental conditions for preparing TiO2 P25 for toxicity testing in A549 cells. Results showed that the use of fetal bovine serum to maintain sufficient colloid stability of dispersed TiO2 P25 is crucial. Under these conditions, TiO2 P25 did not significantly impair A549 cells. When the defined parameters for using TiO2 P25 in A549 cells were met, similar biological effects results were obtained in two independent cell laboratories.

Freeze-thawing impairs the motility, plasma membrane integrity and mitochondria function of boar spermatozoa through generating excessive ROS

Bin Zhang et al.

Summary: In this study, the quality of boar spermatozoa at different steps of cryopreservation was evaluated, revealing that excessive ROS during freeze-thaw cycling can negatively affect sperm motility, plasma membrane integrity, mitochondrial activity, sperm chromatin structure, and apoptosis. Addition of the antioxidant NAC reversed these negative effects, highlighting the importance of ROS downregulation for successful frozen pig sperm production and livestock propagation.

Current Approaches of Preservation of Cells During (freeze-) Drying

Ute Rockinger et al.

Summary: The widespread application of therapeutic cells requires successful stabilization for the duration of transport and storage. Cryopreservation is considered the gold standard for cell storage, but (freeze-) drying could offer higher stability at ambient temperatures. Various excipients, classified into sugars, macromolecules, polyols, antioxidants and chelating agents, are used to prevent cell damage during the (freeze-) drying process.
