Arpita Ghosh et al.

Summary: The study identified three conserved and stable rG4 structures in the 3' region of MALAT1 that regulate alternative splicing, and demonstrated the importance of rG4s in the localization of NCL and NPM to nuclear speckles. Disruption of rG4 in MALAT1 resulted in altered pre-mRNA splicing of endogenous genes, similar to NCL knockdown.

Small molecule quercetin binds MALAT1 triplex and modulates its cellular function

Isha Rakheja et al.

Summary: This study demonstrates that quercetin binds strongly to the triple-helix structure of metastasis-associated lung adenocarcinoma transcript 1 (MALAT1) and modulates its functions. The binding of quercetin to MALAT1 is a spontaneous interaction with a stoichiometry of 1:1 and a negative change in free energy. Furthermore, quercetin downregulates the transcript levels of MALAT1 and reduces its expression in nuclear speckles, leading to modulation of downstream genes.

Dynamics and functional roles of splicing factor autoregulation

Fangyuan Ding et al.

Summary: A study has found that negative autoregulatory splicing of the splicing factor SRSF1 plays a critical role in gene regulation. It establishes an upper limit for SRSF1 protein concentration, reduces cell-to-cell variability in SRSF1 levels, and buffers transcriptional variation. Furthermore, this negative autoregulatory splicing adapts SRSF1 splicing activity to variations in demand from other pre-mRNA substrates.

hnRNPLL controls pluripotency exit of embryonic stem cells by modulating alternative splicing of Tbx3 and Bptf

Xue Wang et al.

Summary: The study reveals the critical role of RNA-binding proteins in pluripotency exit of embryonic stem cells, with hnRNPLL being identified as a key regulator of alternative splicing in ES cells. Its depletion leads to sustained expression of ES cell-preferred isoforms, resulting in differentiation deficiency and developmental defects. This highlights the potential application of targeting RBPs in controlling ES cell fate.

A genome-scale CRISPR screen reveals factors regulating Wnt-dependent renewal of mouse gastric epithelial cells

Kazuhiro Murakami et al.

Summary: Safe utilization of the regenerative potential of adult stem cells for clinical applications requires a comprehensive understanding of the regulatory mechanisms. Epithelial organoid cultures accurately mimic in vivo stem cell-driven epithelial renewal, offering a great ex vivo platform for studying key regulatory mechanisms. Through genome-scale CRISPR knockout screening, Alk, Bclaf3, and Prkra were identified as potential regulators of gastric epithelial differentiation and could suppress stemness/proliferation.

SRSF1 serves as a critical posttranscriptional regulator at the late stage of thymocyte development

Zhihong Qi et al.

Summary: This study reveals a crucial role of serine/arginine-rich splicing factor 1 (SRSF1) in regulating thymocyte maturation by controlling gene networks involved in differentiation, proliferation, apoptosis, and interferon signaling pathway. Deletion of SRSF1 results in severe defects in thymocyte development, affecting the maturation and migration of T cells in the thymus.

Long noncoding RNA DGCR5 involves in tumorigenesis of esophageal squamous cell carcinoma via SRSF1-mediated alternative splicing of Mcl-1

Yuqing Duan et al.

Summary: The study reveals that upregulated lncRNA DGCR5 promotes proliferation, migration, and invasion of ESCC cells while inhibiting apoptosis. DGCR5 directly binds to SRSF1, increasing its stability and initiating important isoform switch of Mcl-1. Cell-derived xenograft model confirms DGCR5's role in facilitating ESCC tumorigenesis.

Nucleo-cytoplasmic shuttling of splicing factor SRSF1 is required for development and cilia function

Fiona Haward et al.

Summary: Shuttling RNA-binding proteins play a crucial role in coordinating gene expression between the nucleus and cytoplasm. By creating a mouse model with SRSF1 protein exclusively retained in the nucleus, researchers found that this alteration led to various cellular and tissue defects, particularly in the context of ciliogenesis. This study highlights the importance of SRSF1 shuttling in reprogramming gene expression networks under high cellular demands, such as during motile ciliogenesis.
