The physiology of alternative splicing

Single-cell architecture and functional requirement of alternative splicing during hematopoietic stem cell formation

Summary: This study constructed a transcriptional atlas of mouse endothelial-to-hematopoietic stem cell transition based on isoforms using single-cell full-length transcriptome data, and revealed the important role of RNA alternative splicing in hematopoietic function. The results showed that early deficiency of Srsf2 in endothelial cells affected the splicing pattern of key hematopoietic regulators and significantly impaired HSC generation.

Protein network analysis revealed the interactions of the LCN2-SLC22A17-MMP9 network with fibronectin 1 and claudin 7 in the TME. Nonsense-mediated RNA decay: an emerging modulator of malignancy

Summary: Nonsense-mediated RNA decay (NMD) is a highly conserved RNA turnover pathway that selectively degrades RNAs harboring truncating mutations, which can affect the expression of tumor suppressor genes and oncogenes. NMD may have a dual role in tumors, as it can promote tumor growth by downregulating the expression of tumor suppressor genes and degrade immunogenic neoantigen peptides, but it can also suppress tumor generation and progression by degrading specific normal mRNAs and non-coding RNAs beneficial for tumors. NMD-modulatory therapy has the potential to provide widespread therapeutic benefit against diverse tumor types, but careful analysis is needed to determine whether NMD should be stimulated or repressed in specific tumors.
