Universal chimeric Fcy receptor T cells with appropriate affinity for IgG1 antibody exhibit optimal antitumor efficacy

Wen Zhu et al.

Summary: There is an urgent need to develop universal CARs with improved flexible targeting and controllable activities. This study systematically investigated the impact of Fcy receptor (FcyR) affinity on CAR-T cells properties by constructing universal CARs using Fcy receptors with different affinities for IgG1 antibodies. The results demonstrated that redirecting and regulating the activities of universal CAR-T cells on tumor cells could be achieved through IgG1 antibodies, and only the FcyRCAR with an appropriate affinity can offer the optimal antitumor advantages of CAR-T cells.

Activation-induced cell death in CAR-T cell therapy

Tian Huan et al.

Summary: CAR-T cells have shown remarkable therapeutic effects on hematologic malignancies, but less effective on solid tumors due to weak persistence. Understanding the mechanism of AICD is crucial to improve CAR-T cell therapy against solid tumors.

Tumor-reactive antibodies evolve from non-binding and autoreactive precursors

Roei D. Mazor et al.

Summary: This study found that somatic hypermutations (SHMs) promote antibody antitumor reactivity against surface autoantigens in high-grade serous ovarian carcinoma (HGSOC). The tumor-reactive antibodies target MMP14, which is abundantly expressed on the tumor cell surface. These findings highlight the origin and potential applicability of autoantibodies directed at surface antigens for tumor targeting in cancer patients.

Human CD19-specific switchable CAR T-cells are efficacious as constitutively active CAR T-cells but cause less morbidity in a mouse model of human CD19+ malignancy

Christopher A. Pennell et al.

Summary: Current FDA-approved CAR-T cell therapies for B-cell malignancies are effective but can increase morbidity and mortality. This study compared the performances of regulatable and constitutively active CAR-T cells, and found that regulatable CAR-T cells have the potential for improved efficacy and reduced toxicity.

Mitochondrial Metabolism Regulation of T Cell-Mediated Immunity

Elizabeth M. Steinert et al.

Summary: Recent evidence suggests that mitochondrial metabolism is crucial for T cell activation, proliferation, and function by providing key metabolites for macromolecule synthesis and generating metabolites necessary for T cell function. This review focuses on how mitochondrial metabolism regulates the fate and function of both conventional and regulatory T cells.

Transient rest restores functionality in exhausted CAR-T cells through epigenetic remodeling

Evan W. Weber et al.

Summary: This study showed that transient cessation of chimeric antigen receptor (CAR) signaling can enhance the efficacy of CAR-T cells, reactivating their anti-tumor functions, and challenging the notion that exhaustion is an epigenetically fixed state.

Barriers to antibody therapy in solid tumors, and their solutions

Yasuhiro Matsumura

Summary: Antibody drugs have become mainstream in cancer treatment, but several barriers such as resistance mechanisms and solid tumor microenvironment features hinder their delivery. A new strategy targeting cancer stroma is proposed, suggesting a shift in the concept of antibody drug development towards cancer-specific targeted therapies.
