Adult hippocampal neu in Alzheimer's disease : A roadmap to clinical relevance

Evgenia Salta et al.

Summary: Adult hippocampal neurogenesis (AHN) decreases significantly in early stages of Alzheimer's disease (AD), but the underlying mechanisms remain unknown. AHN is correlated with cognitive status in AD patients, and understanding its regulation could lead to innovative pharmacological interventions. This study combines molecular, behavioral, and clinical data to explore the complex relationship between AHN and AD pathophysiology and proposes a roadmap for further research to better understand the role of AHN in AD.

Schizophrenia is defined by cell-specific neuropathology and multiple neurodevelopmental mechanisms in patient-derived cerebral organoids

Michael Notaras et al.

Summary: This study utilized patient-derived induced pluripotent stem cells to generate 3D cerebral organoids to model neuropathology of Schizophrenia. The results revealed specific alterations in neuronal programming factors, disrupted neurogenesis, and changes in growth factor expression, suggesting that Schizophrenia neuropathology could be encoded on a cell-type-by-cell-type basis. Single-cell sequencing identified the depletion of BRN2 and PTN, which were found to be mechanistic substrates of neurogenesis and cellular survival in Schizophrenia organoids.

Neurogenesis in aging and age-related neurodegenerative diseases

Luka Culig et al.

Summary: Adult neurogenesis is a potential target for extending cognitive healthspan, as aging is a major risk factor for neurodegenerative diseases. This review describes the role of adult neurogenesis in neurodegenerative diseases and discusses the molecular mechanisms involving key proteins. Interventions that increase neurogenesis and regulate aging research targets are summarized, and the outlook for restoring neurogenesis levels in elderly individuals and those with neurodegeneration is shared.

WDR81 regulates adult hippocampal neurogenesis through endosomal SARA-TGFβ signaling

Min Wang et al.

Summary: Adult hippocampal neurogenesis, crucial for hippocampal function, is regulated by the gene WDR81, mutations of which are linked to neurological disorders. Ablation of WDR81 in adult neural progenitor cells decreases neurogenesis by suppressing endosomal PtdIns3P synthesis. The defective neurogenesis in WDR81-deficient mice can be ameliorated by inhibiting PI3K-III activity or suppressing SARA-dependent TGF beta signaling.

Neural Stem Cell Niche and Adult Neurogenesis

Yue Li et al.

Summary: Adult neurogenesis is a crucial process that generates new and functional neurons from neural stem cells throughout life. Maintaining the proper function of the neurogenic niche requires niche cells to respond to external cues and adapt their roles to influence NSC behavior. Understanding the interactive regulation of NSCs and neurogenesis by the neurogenic niche is important for maintaining the integrative process and functional integrity of the adult brain.

Restoring miR-132 expression rescues adult hippocampal neurogenesis and memory deficits in Alzheimer's disease

Hannah Walgrave et al.

Summary: Neural stem cells in the hippocampal neurogenic niche play a crucial role in lifelong neurogenesis, while miR-132 has been identified as a significant regulator affecting hippocampal neurogenesis in Alzheimer's disease. Research demonstrates that replacing miR-132 can restore neurogenesis and memory deficits in the hippocampus of AD mice, highlighting the potential therapeutic efficacy of targeting miR-132 in neurodegenerative diseases.

Impact of neurodegenerative diseases on human adult hippocampal neurogenesis

J. Terreros-Roncal et al.

Summary: Disrupted hippocampal performance is a major factor contributing to psychiatric comorbidities and cognitive impairments in patients with neurodegenerative disorders. Adult hippocampal neurogenesis (AHN) plays a crucial role in several neurodegenerative diseases, with abnormalities found in adult-born dentate granule cells. Aging and diseases alter the neurogenic niche, reduce microglial phagocytic capacity, trigger astrogliosis, and change the microvasculature of the dentate gyrus, leading to enhanced vulnerability of AHN to neurodegeneration.

Enhancing myelin renewal reverses cognitive dysfunction in a murine model of Alzheimer's disease

Jing-Fei Chen et al.

Summary: The study found a significant increase in new myelin formation rate in the APP/PS1 mouse model of Alzheimer's disease, despite a decrease in overall myelination levels in the cortex and hippocampus. Enhancing myelin renewal through genetic or pharmacological methods improved cognitive performance in memory-related tasks.
