2D Nanozymes Modulate Gut Microbiota and T-Cell Differentiation for Inflammatory Bowel Disease Management

The involvement of TH17 cells in the pathogenesis of IBD

Summary: The pathogenesis of inflammatory bowel disease (IBD) is still unclear and immune dysfunction might play a key role. CD4+ T helper (Th) cells are especially important in this process, with Th17 cells being a major component of CD4+ T cells. Th17 cells produce IL-17 and protect mucous membranes and epithelial tissues against infection. However, when immune regulation is dysfunctional, Th17 cells proliferate abnormally and produce large amounts of proinflammatory cytokines, leading to abnormal immune responses and the development of autoimmune diseases. Recent studies have shown the importance of Th17 cells in the pathogenesis of IBD and their potential as a target for therapy.

Summary: The disruption of mechanical and immune barriers is a major factor in the aggravation of inflammatory bowel disease (IBD). A multifunctional nanozyme capable of scavenging reactive oxygen species (ROS) and inhibiting ferroptosis or T cell differentiation has been developed for IBD therapy. The nanozyme, called Se-HMPB, can effectively scavenge ROS and inhibit ferroptosis in inflammatory tissues, while also regulating T cell differentiation in IBD models. This suggests great potential for alleviating IBD.

An oral nano-antioxidant for targeted treatment of inflammatory bowel disease by regulating macrophage polarization and inhibiting ferroptosis of intestinal cells

Summary: Inflammatory bowel disease (IBD) is a chronic inflammatory disease characterized by excessive reactive oxygen species (ROS) and high ferroptosis. The developed oral nano-antioxidant (Ce-Cur@MCS) targets macrophages in colonic inflammation, mimicking the functions of SOD and CAT to eliminate ROS and protect intestinal cells from ferroptosis. In vivo experiments confirm that the nano-antioxidant enhances therapeutic efficacy of IBD by suppressing macrophage-induced inflammation and inhibiting ferroptosis of intestinal cells. Additionally, it increases the abundance and diversity of gut microflora to maintain therapeutic efficacy of IBD.

Design of Diselenide-Bridged Hyaluronic Acid Nano-antioxidant for Efficient ROS Scavenging to Relieve Colitis

This study developed a nanoformulation, called diselenide-bridged hyaluronic acid nanogel (SeNG), which specifically accumulated in colitis tissue and showed significant anti-inflammatory effects in a mouse model of colitis.
