GSH and H2O2 dynamic correlation in the ferroptosis pathways revealed by engineered probe in tumor and kidney injury

Yuting Wang et al.

Summary: Ferroptosis, a type of cell death caused by lipid peroxidation, is closely related to the development of tumors, ischemia-reperfusion injury, and other diseases. It involves the accumulation of reactive oxygen species and thiols, which promote or inhibit ferroptosis. Understanding the dynamic correlation between reactive oxygen species and active sulfur in these pathways is crucial for revealing the regulation mechanism of ferroptosis.

Peroxynitrite/Lipid Droplet Sequence-Activated Dual-Lock Fluorescent Probes Enable Precise Intraoperative Imaging of Atherosclerotic Plaques

Mangmang Sang et al.

Summary: In this study, a dual-analyte sequentially activated logic fluorescence reporting system CNN2-B was introduced to precisely identify atherosclerotic plaques in vivo. This probe was able to detect peroxynitrite and lipid droplets, the two hallmarks of atherosclerosis, and exhibited higher selectivity and sensitivity than the commercial probe hydroxyphenyl fluorescein (HPF). Fluorescence imaging using CNN2-B clearly delineated atherosclerotic plaques in mouse models.

Ferroptosis of tumour neutrophils causes immune suppression in cancer

Rina Kim et al.

Summary: Ferroptosis is a unique and targetable immunosuppressive mechanism in the tumor microenvironment, which can be pharmacologically modulated to limit tumor progression. It induces spontaneous death of PMN-MDSCs in the tumor microenvironment and limits the activity of human and mouse T cells.

Development of novel near-infrared GFP chromophore-based fluorescent probes for imaging of amyloid-? plaque and viscosity

Yuhui Guo et al.

Summary: Alzheimer's disease is closely associated with abnormal deposition of amyloid beta peptides and plaque formation. In this study, researchers developed a fluorescent probe with improved structure to achieve high selectivity for imaging Alzheimer's plaques in the near-infrared window. The probe showed high fluorescence signal-to-noise ratio and selectivity in living animals and cells, making it an effective tool for monitoring the pathological process of Alzheimer's disease.

Ferroptosis: a cell death connecting oxidative stress, inflammation and cardiovascular diseases

Yi Yu et al.

Summary: Ferroptosis is a newly identified iron-dependent cell death mechanism that differs from other cell death pathways, characterized by lipid peroxidation and accumulation of redox-active iron. Studies suggest that ferroptosis plays a crucial role in various cardiovascular diseases, and targeting ferroptosis may present a novel therapeutic approach in treating these conditions.

Lipid Metabolism in Ferroptosis

Zhi Lin et al.

Summary: Lipid metabolism plays a crucial role in regulating cell survival and death. Abnormal lipid metabolism can lead to ferroptosis, an iron-dependent regulated cell death. Various antioxidant systems and membrane repair pathways help mitigate oxidative damage and support cell survival, especially in pathological conditions like cancer. Impairment of the ferroptosis machinery is associated with the development of diseases such as cancer and tissue damage.

Lipid Droplet-Specific Probe for Rapidly Locating Atherosclerotic Plaques and Intraoperative Imaging via In Situ Spraying

Mangmang Sang et al.

Summary: The study introduces an aerosol spraying technique of a lipid droplet-specific probe to precisely locate atherosclerotic plaques during surgery. This method allows for stable imaging of the plaque within 5 minutes, accurately delineating the plaque margins and size, with weak interference from background signals. The intraoperative fluorescence imaging via in situ delivery of the lipid droplet-specific probe shows promise for effective clinical application.

Development of Near-Infrared Nucleic Acid Mimics of Fluorescent Proteins for In Vivo Imaging of Viral RNA with Turn-On Fluorescence

Jiaheng Zhang et al.

Summary: By structurally modulating GFP-like chromophores using a specific molecular configuration, researchers have created novel fluorogenic derivatives with infrared-shifted spectra. These derivatives can be fluorescently elicited by interacting with G-quadruplex structures, leading to the creation of FP mimics with tunable emission in the near-infrared region.

Summary: In recent years, coumarin fluorescent probes have been widely used in various fields due to their excellent optical properties. This study successfully prepared two probes with high quantum yield and rapid response by introducing electron-donating group and pyrazoline ring at different positions, providing a new concept for the construction and optimization of fluorescent probes. The probe exhibited synergistic action of -OH and pyrazoline rings, effectively detecting ClO- in cells and zebrafish.

Twisted intramolecular charge transfer (TICT) and twists beyond TICT: from mechanisms to rational designs of bright and sensitive fluorophores

Chao Wang et al.

Summary: This review highlights the importance of the TICT mechanism in guiding the development of bright and sensitive fluorophores. It also summarizes the molecular design strategies and compares the twisted intramolecular charge shuttle (TICS) and twists during photoinduced electron transfer (PET) mechanisms. Understanding the structure-property relationships between twisted geometries and fluorescence can aid in the rational development of fluorophores involving molecular twisting in the excited state.
