Acceleration of pelvic tissue generation by overexpression of basic fibroblast growth factor in stem cells

Summary: This study aimed to address issues related to growth factors application, long-term exogenous treatment side effects, and stem cell directional differentiation. Evaluation of the paracrine effects and directional differentiation of adipose mesenchymal stem cells through stable overexpression of basic fibroblast growth factor (bFGF) showed that modified stem cells could continuously release bFGF and spontaneously differentiate into fibroblasts with high efficiency.

Summary: The driving factors for fibrosis and scar formation are fibroblast differentiation and hampered myofibroblast apoptosis. This study developed a high-throughput in vitro fibrosis model using synthetic polyisocyanide hydrogels, which allowed for measuring contraction and subsequent molecular and cellular analysis. The model successfully differentiated fibroblasts and myofibroblasts and was used to test the effects of antifibrotic drugs.

Novel Thermosensitive Hydrogel Promotes Spinal Cord Repair by Regulating Mitochondrial Function

Summary: This study presents a promising bioactive system for the treatment of spinal cord injury (SCI) by immobilizing umbilical cord mesenchymal stem cells (UCMSC) and basic fibroblast growth factor (bFGF) in extracellular matrix (ECM) and heparin-poloxamer (HP). The results show that this system has therapeutic effects in reducing apoptosis and improving mitochondrial function, and it activates PAK1 and SIRT4 to protect against SCI.

Polyisocyanides as a substrate to trigger vaginal fibroblast functioning in an in vitro model for prolapse repair

Summary: Pelvic organ prolapse (POP) is a condition where the bladder, uterus, and/or rectum descend into the vagina. It is associated with changes in vaginal fibroblast functionality and connective tissue composition. Surgical intervention for POP has poor outcomes, possibly due to the lack of true connective tissue restoration. A novel treatment approach involves using a bioactive supplement to enhance tissue repair post-surgery. In this study, the effects of introducing cell binding peptides on a biomimetic gel were investigated using primary vaginal fibroblasts from a POP patient. The results showed that the modified gel promoted cell spread and proliferation, and led to increased production of collagen and elastin by the fibroblasts. These findings suggest that this gel application may improve connective tissue healing in the pelvic floor, which has important implications for regeneration in surgery.
