NIR-II Regulation of Mitochondrial Potassium Channel with Dual-Targeted Conjugated Oligomer Nanoparticles for Efficient Cancer Theranostics In Vivo

Anti-Quenching NIR-II J-Aggregates of Benzo[c]thiophene Fluorophore for Highly Efficient Bioimaging and Phototheranostics

Summary: A bright benzo[c]thiophene (BT) J-aggregate fluorophore (BT6) with anti-quenching effect is reported for highly efficient NIR-II bioimaging and phototheranostics. The BT fluorophores have Stokes shift over 400 nm and aggregation-induced emission (AIE) property, overcoming the self-quenching issue of J-type fluorophores. In an aqueous environment, BT6 assemblies exhibit absorption over 800 nm and NIR-II emission over 1000 nm, resulting in enhanced imaging capabilities. The BT6 NPs show excellent performance in NIR-II fluorescence imaging and cancer phototheranostics, as demonstrated by in vivo visualization and imaging-guided phototherapy results.

Controlled Activation of TRPV1 Channels on Microglia to Boost Their Autophagy for Clearance of Alpha-Synuclein and Enhance Therapy of Parkinson's Disease

Summary: This study reports the enhancement of microglial autophagy through the phagocytosis and degradation of alpha-synuclein, leading to improved therapeutic efficacy in Parkinson's disease. By using a rationally designed photothermal nanoagent, the surface TRPV1 channels of microglia are controlled, resulting in the activation of autophagy and clearance of alpha-synuclein. In mouse models, this treatment significantly improves motor ability and restores biomarker levels to those of healthy mice.

Near-Infrared Light-Responsive Nanoinhibitors for Tumor Suppression through Targeting and Regulating Anion Channels

Summary: In this study, a natural compound procyanidin (PC) was identified as a selective inhibitor of TMEM16A and its precise electrostatic interactions with TMEM16A were characterized. Near-infrared (NIR)-light-responsive photothermal conjugated polymer nanoparticles encapsulating PC (CPNs-PC) were developed for remote targeting and regulation of TMEM16A. The findings provided a strategy to promote tumor regression by accurately and remotely regulating anion channels using NIR-light-responsive polymer nanoparticles containing specific inhibitors of TMEM16A.

Advances in Prostate-Specific Membrane Antigen (PSMA)-Targeted Phototheranostics of Prostate Cancer

PSMA, as a specific membrane antigen target, is widely expressed in prostate cancer tissues and is correlated with tumor aggressiveness. Phototheranostics, including near-infrared fluorescence imaging, photoacoustic imaging, and photodynamic therapy, have shown great potential for PSMA-targeted treatment and diagnosis. However, there are still challenges and opportunities for the clinical translation of phototheranostics in the field of prostate cancer.

Photothermal Modulation of Depression-Related Ion Channel Function through Conjugated Polymer Nanoparticles

Summary: Through the use of near-infrared light-sensitive nanoparticles, the study demonstrates the potential for modulating the activity of target neurons and developing more precise drug therapies for diseases such as major depression. The nanoparticles show excellent photothermal effect under NIR irradiation, allowing for effective penetration of the blood-brain barrier and reaching neurons. Loading a selective activator into the nanoparticles results in a remarkable reduction in the firing frequency of dopaminergic neurons, suggesting a new concept for developing selective drug therapies.
