R-spondin-3 is an oncogenic driver of poorly differentiated invasive breast cancer

Eline J. ter Steege et al.

Summary: RSPO2/RSPO3 copy number gains are found in a quarter of breast cancer patients and associated with worse prognosis. A mouse model study reveals the oncogenic capacity of RSPO3 in breast cancer, driving tumor development with poor differentiation, epithelial-to-mesenchymal transition, and metastatic potential. RSPO3 is proposed as a novel candidate target for therapy in breast cancer.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

The role of R-spondin proteins in cancer biology

Eline J. ter Steege et al.

Summary: RSPO proteins play a crucial role in activating the Wnt/beta-catenin signaling pathway in stem cells and progenitor cells. Deregulation of RSPOs has been implicated in various cancer types, with potential for therapeutic targeting in cancer cases with RSPO overrepresentation.

Self-promoted Albumin-Based Nanoparticles for Combination Therapy against Metastatic Breast Cancer via a Hyperthermia-Induced Platelet Bridge

Wei Zhao et al.

Summary: This study developed a photothermal therapy strategy using activated platelets as bridges to enhance drug delivery efficacy for inhibiting the growth of primary tumors and metastasis in metastatic cancer treatment. By utilizing mitochondrial targeting, platelet targeting, and metastasis-EPR effects, the constructed nanoplatform effectively improved drug accumulation and inhibited tumor growth and metastasis.

Apc-mutant cells act as supercompetitors in intestinal tumour initiation

Sanne M. van Neerven et al.

Summary: In the ISC niche, the delicate balance of WNT agonists and antagonists is crucial for maintaining the ISC compartment and preventing unrestrained activation of the WNT pathway. Apc-mutant ISCs function as supercompetitors by secreting WNT antagonists to induce differentiation of neighboring wild-type ISCs. Lithium chloride prevents the expansion of Apc-mutant clones and adenoma formation by rendering wild-type ISCs insensitive to WNT antagonists through downstream activation of WNT by inhibiting GSK3 beta. Boosting the fitness of healthy cells may be a powerful strategy to limit the formation of cancers in high-risk individuals.
