The pGinger Family of Expression Plasmids

Summary: The pGinger suite of expression plasmids consists of 43 plasmids that allow precise constitutive and inducible gene expression in various Gram-negative bacteria. These plasmids include 16 synthetic constitutive promoters upstream of red fluorescent protein (RFP), with a wide range of hosts and antibiotic resistance markers. The family also contains seven inducible systems controlling RFP expression, with variants that utilize different origins and selection markers. Relevant data have been collected in Escherichia coli and Pseudomonas putida, and all pGinger vectors are available through the JBEI Public Registry.

Improved polyketide production in C. glutamicum by preventing propionate-induced growth inhibition

Summary: Corynebacterium glutamicum is a promising host for producing valuable polyketides, but propionate addition inhibits its growth. This study identifies the accumulation of propionyl-CoA and methylmalonyl-CoA as the cause of growth inhibition and demonstrates that it can be relieved by introducing methylmalonyl-CoA-dependent polyketide synthases. Laboratory evolution is used to increase polyketide production by targeting mutations in germicidin synthase and citrate synthase. These findings highlight the importance and potential of C. glutamicum in polyketide production.

Genome-wide role of codon usage on transcription and identification of potential regulators

Summary: Codon usage bias plays a critical role in regulating gene expression levels, not only through translation but also through transcription in fungal and mammalian genes. In Neurospora, the correlation between codon usage and RNA levels is maintained both genome-wide and in the nucleus, suggesting a broad influence of codon usage on mRNA levels through transcription in a translation-independent manner. A large-scale genetic screen identified multiple chromatin regulators and transcription factors as mediators of the transcriptional effect of codon usage.
