Sensory Perception in Bacterial Cyclic Diguanylate Signal Transduction

Summary: Cyclic diguanylate (c-di-GMP) signal transduction systems allow bacteria to sense and respond to changing cell status or environmental conditions. This review provides a comprehensive overview of the stimuli and receptors that modulate intracellular c-di-GMP levels. The analysis of sensory domains in diguanylate cyclases and c-di-GMP-specific phosphodiesterases, as well as their associated receptor complexes, reveals a diverse repertoire of protein domain families involved in these functions. The principles of stimulus perception learned from studying these modular sensory devices are described, along with their assembly in various combinations with output domains. Furthermore, a system for classifying these sensor proteins based on their complexity is summarized. Biological information processing via c-di-GMP signal transduction is not only crucial for bacterial survival in dynamic environments, but also holds potential for engineering gene expression circuitry and synthetic proteins with customized biochemical functionalities.

Summary: Proteins can have different structures, ranging from rigid to flexible to completely disordered. Intrinsic disorder proteins (IDPs) or specific intrinsically disordered regions (IDRs) can form complexes that would be otherwise impossible, providing the flexibility needed for specific functions. IDRs can bind multiple partners and act as hubs in protein-protein interaction networks. The disorder-to-order transition during binding provides specificity and reversibility. The interactions of IDRs can be modulated by the environment or covalent modifications, and their mis-signalling or non-native folding may lead to diseases.

Cyclic di-GMP Regulates the Type III Secretion System and Virulence in Bordetella bronchiseptica

Summary: This study investigates the role of c-di-GMP in Bordetella bronchiseptica using a multi-omics approach. The results show that c-di-GMP regulates biofilm formation, swimming motility, and the expression of virulence factors in this bacterium. This inhibition of T cell response by aPV may have led to the resurgence of pertussis by augmenting the Bp reservoir and hindering neutrophil recruitment essential for clearing the bacteria.

Summary: Bordetella bronchiseptica is a common cause of upper respiratory tract infections in domesticated dogs and cats, and a rare zoonotic pathogen in immunocompromised humans. In patients with HIV/AIDS, this infection can lead to pneumonia with interstitial infiltrates, often accompanied by sepsis. Advanced immune suppression and other chronic conditions that compromise host defenses are commonly associated with B bronchiseptica infection in patients who do not have HIV infection.
