The role of proteolysis in interleukin-11 signaling

Juliane Lokau et al.

Summary: IL-11, a member of the IL-6 family of cytokines, was originally discovered as a factor that promotes megakaryocyte maturation, but recent research has revealed its crucial roles in inflammation, fibrosis, and cancer. IL-11 initiates signaling through both classic and trans-signaling mechanisms, impacting the activation of cells that do not express the IL-11 receptor.

The outcome of targeted NGS screening in patients with syndromic forms of sagittal and pansynostosis-IL11RA is an emerging core-gene for pansynostosis

Alexandra Topa et al.

Summary: In this study, the prevalence and spectrum of genetic alterations in syndromic forms of sagittal and pansynostosis were investigated. Pathogenic and likely pathogenic variants were found in 72% of the cases, with FGFR2, TWIST1, IL11RA, and SKI being the most affected genes. Two novel likely pathogenic variants in FGFR2 and TWIST1, as well as a novel variant of unclear significance in RUNX2, were identified. Notably, three new patients with pansynostosis and an IL11RA mutation were also identified. Targeted NGS using a broad panel of CS-related genes proved to be a simple and powerful tool for detecting pathogenic mutations in patients with syndromic forms of CS.

Interleukin-11 (IL-11) receptor cleavage by the rhomboid protease RHBDL2 induces IL-11 trans-signaling

Lydia Koch et al.

Summary: Interleukin-11 (IL-11) activates target cells through different signaling pathways, including the shedding of IL-11R triggered by RHBDL2. RHBDL2 can cleave IL-11R within the cell, generating biologically active sIL-11R capable of trans-signaling IL-11 action.

Similarities and differences between IL11 and IL11RA1 knockout mice for lung fibro-inflammation, fertility and craniosynostosis

Benjamin Ng et al.

Summary: The loss of function in IL11 signaling is crucial for fertility, fibrosis, inflammation, and craniosynostosis. Studies on IL11 knockout mice show resistance to pro-fibrotic stimulation and protection from pulmonary fibrosis. IL11 may play a role in bone development abnormalities associated with IL11RA mutation.

A Novel Missense Mutation in TNNI3K Causes Recessively Inherited Cardiac Conduction Disease in a Consanguineous Pakistani Family

Shafaq Ramzan et al.

Summary: The study investigated a consanguineous Pakistani family with four patients with cardiac conduction disease and identified a novel homozygous missense mutation in the TNNI3K gene. Molecular dynamics simulations showed changes in protein surface and hydrogen bond network due to the mutation. Analysis revealed structural variations in the ATP-binding pocket and homodimer interface caused by the mutation, suggesting it as a pathogenic variant.
