Diversity of the bacteriocins, their classification and potential applications in combat of antibiotic resistant and clinically relevant pathogens

Gee-Hyeun Choi et al.

Summary: It has been almost a century since the discovery of penicillin, and during this time, there has been great enthusiasm and abuse in the development and use of antibiotics. However, the emergence of antibiotic resistance has led to the realization that new types of antimicrobials are needed. Bacteriocins, produced by various types of bacteria and other organisms, have been explored as potential antimicrobials, particularly in the food industry. In recent decades, bacteriocins have shown promise as alternative therapies for treating antibiotic-resistant pathogens. They are highly selective antimicrobials with low cytotoxicity. The application of natural or bioengineered bacteriocins, along with synergistic combinations of bacteriocins and conventional antibiotics, may be the next step in combating drug-resistant pathogens. This overview focuses on the diversity of lactic acid bacteria and their bacteriocins, but also discusses examples from non-lactic acid bacteria, archaea, and eukaryotes. The therapeutic properties, bioengineering, and combined applications of bacteriocins with conventional antibiotics are evaluated for their potential in human and veterinary medicine.

Association of Kidney Stones and Recurrent UTIs: the Chicken and Egg Situation. A Systematic Review of Literature

Francesco Ripa et al.

Summary: This review examines the association between kidney stone disease (KSD) and recurrent urinary tract infections (rUTI), and assesses the outcomes of kidney stone treatment on resolving rUTI. The review finds a strong association between KSD and rUTI, with surgical clearance of kidney stones usually resulting in resolution of UTI. However, there is discordant data on recurrence rates after surgery. In vitro studies suggest a possible causative role of bacteria in metabolic stone formation. The review recommends proactive intervention through stone removal in patients with recurrent UTIs or additional risk factors, and tailoring antibiotic treatment based on stone culture results.

The extracellular domain of site-2-metalloprotease RseP is important for sensitivity to bacteriocin EntK1

Sofie S. Kristensen et al.

Summary: This study investigates the molecular mechanisms underlying the interaction between RseP and Enterocin K1 (EntK1) and the bacteriocin sensitivity. Through mutational analysis, the researchers identified key regions and motifs of RseP involved in bacteriocin binding and activity. They found that the extracellular PDZ domain is the main determinant of EntK1 sensitivity. These findings may contribute to the development of novel bacteriocins and the identification of RseP as a potential antibacterial target.

Bacteriocins: Potential for Human Health

Fuqing Huang et al.

Summary: Bacteriocins have gained attention as antimicrobial compounds due to the challenges of antibiotic resistance, with potential applications in food preservation and as clinical antimicrobials or immune-modulating agents. They have broad- or narrow-spectrum antimicrobial activity against clinically pathogenic and multidrug-resistant bacteria, and also show anticancer, anti-inflammatory, and immune-modulatory activities. Their safety, low likelihood of causing drug resistance, better efficacy, and application prospects make them promising therapeutic agents.

A bacteriocin-based treatment option for Staphylococcus haemolyticus biofilms

Christian Kranjec et al.

Summary: A novel hybrid bacteriocin H1 showed superior antimicrobial effects against Staphylococcus haemolyticus and high activity towards various strains without strain-dependency. Combination with broad-spectrum bacteriocins micrococcin P1 and garvicin KS effectively eradicated biofilm-associated cells and prevented development of resistant mutants, indicating its potential as a treatment option for S. haemolyticus.

Trend of Bacterial Uropathogens and Their Susceptibility Pattern: Study of Single Academic High-Volume Center in Italy (2015-2019)

Enrica Serretiello et al.

Summary: Urinary tract infections (UTIs) are a common infection that can affect individuals of different age groups, genders, and pregnancy/menopause states. This study conducted at a university hospital in Italy provides data on UTI incidence, gender distribution, age groups affected, and antibiotic resistance profiles of the most representative bacterial strains. The findings reveal a higher incidence of UTIs in the elderly population, with Gram-negative bacteria and Candida species being commonly identified pathogens.

Bacteriocins as a new generation of antimicrobials: toxicity aspects and regulations

Samira Soltani et al.

Summary: Bacteriocins have gained attention as antimicrobial compounds in recent decades, with potential applications in clinical settings and as immune-modulating agents. Despite their effectiveness against antibiotic-resistant bacteria, limited safety and toxicity data hinder their broader use in the food and medical industry. Nisin remains the only bacteriocin approved as a food preservative by regulatory agencies.
