The C-terminal tail of α-synuclein protects against aggregate replication but is critical for oligomerization

Azad Farzadfard et al.

Summary: Farzadfard et al. conducted a comprehensive analysis on a range of C-terminal truncations of aSN, revealing the importance of high C-terminus charge for decreased fibrillation rates. Truncated aSN showed decreased ability to form oligomers, disrupt synthetic vesicles, and exhibit cell toxicity, while still promoting or inhibiting aggregation through intramolecular interactions. Truncation of the C-terminal domain not only increased fibrillation tendency but also suppressed toxic oligomer formation, with changes in electrostatic potential serving as a double-edged safety guard.

Mechanisms of enhanced aggregation and fibril formation of Parkinson's disease-related variants of α-synuclein

Takashi Ohgita et al.

Summary: The A53T mutation and C-terminal truncation enhance the aggregation of α-synuclein by reducing the unfavorable activation enthalpy of nucleation, and the truncation further triggers the autocatalytic fibril elongation on the fibril surfaces.

Transcellular propagation of fibrillar α-synuclein from enteroendocrine to neuronal cells requires cell-to-cell contact and is Rab35-dependent

Paulla Vieira Rodrigues et al.

Summary: Parkinson's disease is a neurodegenerative disease that can originate in the enteric nervous system and spread to the brain. Enteroendocrine cells in the gut may play a key role in the development of Parkinson's disease. Alpha-synuclein is involved in this process.

A NAC domain mutation (E83Q) unlocks the pathogenicity of human alpha-synuclein and recapitulates its pathological diversity

Senthil T. Kumar et al.

Summary: The E83Q mutation in alpha-synuclein accelerates fibrillization and leads to the formation of fibrils with distinct structural and dynamic properties. In cells, this mutation is associated with increased levels of aSyn, accumulation of p5129, and higher toxicity. In a neuronal seeding model, the E83Q mutation enhances fibril internalization, induces higher seeding activity, and results in the formation of diverse aSyn pathologies.

Pathological Relevance of Post-Translationally Modified Alpha-Synuclein (pSer87, pSer129, nTyr39) in Idiopathic Parkinson's Disease and Multiple System Atrophy

Berkiye Sonustun et al.

Summary: The predominant post-translational modifications of alpha-synuclein in the pathological process of idiopathic Parkinson's disease and multiple system atrophy were found to be phosphorylation and nitration, with phosphorylated alpha-synuclein being the earliest and most abundant form. These findings may have implications for the development of novel biomarkers and therapeutic approaches.

Novel conformation-selective monoclonal antibodies against apoA-I amyloid fibrils

Takashi Ohgita et al.

Summary: Novel monoclonal antibodies (mAbs) generated in this study exhibit strong reactivity to amyloid fibrils formed by the 1-83 fragment of apoA-I, suggesting potential applications in both diagnosis of apoA-I-related amyloidosis and structural analysis of amyloid fibrils. These mAbs preferentially react with mature fibrils over non-fibrillar aggregates and recognize common structural features in apoA-I amyloid fibrils, as well as amyloid fibrils formed by alpha-synuclein.

Structural insights into α-synuclein monomer-fibril interactions

Pratibha Kumari et al.

Summary: This study investigated the interaction between monomeric alpha-Syn and its fibrillar form using NMR and electron paramagnetic resonance spectroscopy, revealing that intermolecular interactions reduce intramolecular contacts in monomeric alpha-Syn, leading to further unfolding of its intrinsically disordered states and critically contributing to the aggregation kinetics of alpha-Syn during secondary nucleation.

Synuclein Regulates Synaptic Vesicle Clustering and Docking at a Vertebrate Synapse

Kaitlyn E. Fouke et al.

Summary: Synuclein plays a key role in regulating the clustering of synaptic vesicles at synapses, with perturbations causing a dose-dependent reduction in the number of vesicles. This protein also affects the docking of vesicles at the active zone, influencing the size and distribution of vesicle clusters. These findings provide insights into the role of synuclein in neurotransmission and its implications for diseases like Parkinson's.

Synthetic NAC 71-82 Peptides Designed to Produce Fibrils with Different Protofilament Interface Contacts

Thomas Nasstrom et al.

Summary: Using molecular dynamics simulations and experimental studies, it was found that double-fragment peptide fibrils have lower relative beta-sheet content compared to single-fragment peptide fibrils. The morphological differences, such as short protofibrils, elongated, twisted, and rod-like structures, in non-capped and capped double-fragment peptide systems were confirmed through TEM analysis. The differences in Thioflavin T fluorescence lifetime profiles upon fibril binding suggest potential morphological differences in the fibril samples.

Amyloid particles facilitate surface-catalyzed cross-seeding by acting as promiscuous nanoparticles

Nadejda Koloteva-Levine et al.

Summary: Amyloid seeds are nanometer-sized protein particles that can accelerate amyloid formation through a surface catalysis mechanism without propagating the specific amyloid conformation associated with the seeds. This allows for cross-seeded assembly reactions involving nonhomologous proteins and rationalizes the molecular mechanism of the cross-seeding phenomenon as a manifestation of aberrant surface activities by amyloid seeds.
