Differential gene expression in the contralateral hemisphere of the rat brain after focal ischemia

Ivan B. Filippenkov et al.

Summary: In this study, functional genetic analysis was performed on the contralateral hemisphere after ischemia-reperfusion injury, revealing complex bilateral reactions at the transcriptome level in the rat brain. It is believed that specific genome responses in the contralateral hemisphere and ischemic hemisphere may serve as a useful model for studying brain repair after stroke.

Unsaturated bond recognition leads to biased signal in a fatty acid receptor

Chunyou Mao et al.

Summary: In this study, six cryo-electron microscopy structures of GPR120 in complex with FA hormones or TUG891 and Gi or Giq trimers were reported. The study also investigated synthetic ligand selectivity and the structural basis of missense single-nucleotide polymorphisms. The findings provide insights into how GPR120 recognizes different double-bond positions of fatty acids and its role in metabolic diseases.

Polyunsaturated fatty acids and fatty acid-derived lipid mediators: Recent advances in the understanding of their biosynthesis, structures, and functions

Simon C. Dyall et al.

Summary: Polyunsaturated fatty acids are important components of cell membranes and play a role in cellular function. They can also serve as precursors for lipid mediators called oxylipins. Oxylipins are formed through enzymatic or non-enzymatic pathways and include eicosanoids and other lipid mediators. This review summarizes current knowledge about the structure and functions of nonclassical oxylipins.

UPR Responsive Genes Manf and Xbp1 in Stroke

Helike Lohelaid et al.

Summary: Stroke is a devastating medical condition with no current treatment to hasten recovery. Boosting endogenous pro-survival pathways through the unfolded protein response (UPR) may provide a potential strategy to improve stroke outcomes, with mesencephalic astrocyte-derived neurotrophic factor (MANF) being identified as a promising UPR responsive gene with neuroprotective effects.

Reactive astrocytes facilitate vascular repair and remodeling after stroke

Michael R. Williamson et al.

Summary: This study demonstrates that reactive astrocytes play a crucial role in vascular repair and remodeling after ischemic stroke in mice, by activating transcriptional programs related to vascular remodeling. Ablation of reactive astrocytes significantly impairs vascular and extracellular matrix remodeling, leading to prolonged blood flow deficits, exacerbated vascular permeability, ongoing cell death, and worsened motor recovery. These findings highlight the critical role of reactive astrocytes as cellular mediators of vascular remodeling during neural repair.

Immune Cells in the BBB Disruption After Acute Ischemic Stroke: Targets for Immune Therapy?

Yan-mei Qiu et al.

Summary: Blood-Brain Barrier (BBB) disruption is a crucial pathophysiological process in acute ischemic stroke, where the rapid activation of immune cells plays a critical role. Infiltrating immune cells increase BBB permeability, causing microvascular disorder, but also promote BBB repair and angiogenesis. Understanding the inflammatory mechanisms behind BBB disruption can lead to the development of more effective therapeutics for acute ischemic stroke.
