Summary: Recent studies have found a genetic link between hearing difficulty and Alzheimer's disease (AD), especially a direct causal link with the APOE gene. Additionally, shared effects of glutamine, gamma-glutamylglutamine, and citrate levels on reducing the risk of both hearing difficulty and AD were observed. Although no statistically significant associations were found after multiple testing corrections, systematic analysis suggests a shared vulnerability in molecular pathways between hearing difficulty and AD.

Summary: This study reassessed the genome-wide significance threshold in GWAS by comparing different multiple testing strategies. The findings suggest that relaxing the P-value threshold can increase discovery rates but also lead to higher false positive rates. FDR and Bayesian FDR are shown to be well controlled for different sample sizes in this context.
