Relationship between Hypoxic and Immune Pathways Activation in the Progression of Neuroinflammation: Role of HIF-1α and Th17 Cells

Summary: Neuroinflammation is a common occurrence in degenerative diseases of both the central and peripheral nervous system, triggered by immune system alterations or inflammatory cascades. The pathophysiology of these disorders involves multiple factors and current therapies have limited clinical efficacy. This review discusses the relationship between deregulation of T helper cells and hypoxia, specifically focusing on the Th17 and HIF-1 alpha molecular pathways, which play a role in the development of neuroinflammation. The clinical presentation of neuroinflammation is seen in prevalent conditions like multiple sclerosis, Guillain-Barre syndrome, and Alzheimer's disease, and potential therapeutic targets related to the pathways involved in neuroinflammation are explored.

Characterization of a novel prolyl hydroxylase 2 gene from mud crab Scylla paramamosain: Insights into its role in the regulation of hypoxia-inducible factor-1 alpha

Summary: In this study, a novel PHD2 gene, SpPHD2, was cloned and identified in the mud crab Scylla paramamosain. The mRNA expression of SpPHD2 was widely distributed across all examined tissues, and the protein was mainly localized in the cytoplasm. Silencing SpPHD2 in vivo upregulated SpHIF-1 alpha and downstream genes, while overexpression of SpPHD2 in vitro dose-dependently reduced SpHIF-1 alpha transcriptional activity, indicating the crucial role of SpPHD2 in regulating SpHIF-1 alpha.

Further research on the relationship between hypoxia and AD may lead to effective preservation and pharmacological treatment of this neurodegenerative disease. This review summarizes the effects of normoxia and hypoxia on AD pathogenesis, discussing the underlying mechanisms and the regulation of HIF-1α and its key players, such as P53, VEGF, and GLUT1.

The Capillary Morphogenesis Gene 2 Triggers the Intracellular Hallmarks of Collagen VI-Related Muscular Dystrophy

Summary: This study reveals the inability of patient-derived fibroblasts to build the extracellular collagen VI network and its correlation with increased phosphorylation of the collagen VI receptor CMG2, leading to the accumulation of endosomes and lysosomes.
