Co-culture models of endothelial cells, macrophages, and vascular smooth muscle cells for the study of the natural history of atherosclerosis

Martin M. Liu et al.

Summary: This study presents a fast, affordable, and reproducible three-cell co-culture system that can represent the different cellular mechanisms of atherosclerosis. The models created in this study closely mimic the behaviors of endothelial cells, macrophages, and vascular smooth muscle cells in human atherosclerotic plaques. These models have the potential to be used for cost- and time-effective investigations of atherosclerosis and new drug therapies.

Dissecting the IL-6 pathway in cardiometabolic disease: A Mendelian randomization study on both IL6 and IL6R

Arjen J. Cupido et al.

Summary: Targeting IL-6 and IL-6 receptor can reduce the risk of multiple cardiometabolic diseases, suggesting both IL-6 and IL-6R are potential therapeutic targets for lowering CVD. Furthermore, IL-6 inhibition may have a more favorable effect on pneumonia risk than IL-6R inhibition.

Inflammation and atherosclerosis: signaling pathways and therapeutic intervention

Peng Kong et al.

Summary: Atherosclerosis is a chronic inflammatory vascular disease that can be promoted or inhibited by innate and adaptive immune responses. Several signaling pathways associated with the inflammatory response have been found to play important roles in the development and regression of atherosclerosis. Targeting inflammatory pathways may represent a new approach for the treatment of atherosclerotic diseases.

Vascular smooth muscle cells in atherosclerosis: time for a re-assessment

Mandy O. J. Grootaert et al.

Summary: Vascular smooth muscle cells play a key role in atherosclerosis by forming a protective fibrous cap and exhibiting various phenotypes that can affect plaque formation and stability. They are a larger proportion of atherosclerotic plaques than previously thought and their plasticity is regulated by various mechanisms.

Role of Interleukin-6 in Vascular Health and Disease

Paulina Villar-Fincheira et al.

Summary: IL-6 is a pleiotropic cytokine involved in regulating immune responses, with dysregulated activity linked to chronic inflammation and autoimmune conditions like atherosclerosis. However, it can also play beneficial roles in situations like exercise, inducing anti-inflammatory effects and metabolic changes. The dual effects of IL-6 may be explained by complex signaling pathways.

Cellular Crosstalk between Endothelial and Smooth Muscle Cells in Vascular Wall Remodeling

Nerea Mendez-Barbero et al.

Summary: Pathological vascular wall remodeling refers to the changes in structure and function of the vessel wall in response to injury, potentially leading to cardiovascular disease. Communication between endothelial cells and vascular smooth muscle cells is crucial for vasculature development and vessel homeostasis, with aberrant communication associated with various disease states including vascular wall remodeling. Different mechanisms such as paracrine regulations and direct contact play a role in this interplay.
