Can Circadian Eating Pattern Adjustments Reduce Risk or Prevent Development of T2D?

Carlee Harris et al.

Summary: Type 2 diabetes (T2D) is caused by insulin resistance and poor diet. High frequency eating disrupts the circadian clock and gut microbiome, leading to T2D. An improved diet with meal consistency, avoiding late-night eating, low meal frequency, and fasting can improve metabolic health and T2D. Early time-restricted eating (TRE) is more beneficial than other fasting methods.

Effects of Intermittent Fasting on Regulation of Metabolic Homeostasis: A Systematic Review and Meta-Analysis in Health and Metabolic-Related Disorders

Ana Ines Silva et al.

Summary: Intermittent fasting (IF) is a dietary intervention associated with improved metabolic parameters. This review and meta-analysis compared different IF protocols, including alternate-day fasting (ADF), time-restricted fasting (TRF), and religious fasting (RF). ADF was found to have the most beneficial effects on metabolic conditions, particularly in obese and MetS individuals. However, the impact of IF on individuals with type 2 diabetes (T2D) was limited, primarily affecting insulin homeostasis. The study highlights the importance of considering an individual's health status and specific metabolic diseases when evaluating the effects of IF.

Multimodal efforts to slow the progression of chronic kidney disease in patients with type 2 diabetes mellitus

Anjay Rastogi et al.

Summary: For patients with chronic kidney disease (CKD) associated with type 2 diabetes mellitus (T2DM), slowing the progression of kidney disease is important. Renin-angiotensin-aldosterone system inhibitors (RAASis) such as ACEIs and ARBs have been shown to have protective effects on both the kidneys and cardiovascular system in CKD patients. Adding an SGLT2 inhibitor to ACEI or ARB therapy provides additional benefits for kidney and cardiovascular protection. In cases of persistent albuminuria despite RAASi therapy, adding a nonsteroidal mineralocorticoid receptor antagonist (finerenone) has shown benefits in clinical trials.

Summary: Diabetes mellitus (D.M.) is a metabolic disorder caused by defects in insulin production and insulin sensitivity. The advancements in genome editing technologies, particularly CRISPR/Cas9, have revolutionized the field of gene editing and enabled targeted modifications in specific cell genome sequences. CRISPR/Cas9 has been effective in ex vivo gene editing in stem cells and the creation of animal models for diabetes, providing insights into pancreatic beta-cell development and evaluating therapeutic strategies.

Niki Mourouti et al.

Summary: The study investigates the association between different lifestyle patterns (LPs) and the existence of prediabetes in adults from families at high risk for type 2 diabetes (T2D) in Europe. The results show that one LP is positively associated with prediabetes, while another LP is not significantly associated with prediabetes. Understanding LPs could provide necessary evidence for planning interventions and education strategies for prediabetes and T2D.

The effects of intermittent fasting and a low-carbohydrate diet on type 2 diabetes

Miri Ann Pelc

Summary: Type 2 diabetes, a complication of obesity, imposes financial burden on the US healthcare system. The 2022 ADA guidelines recommend reducing carbohydrate intake to improve hyperglycemia in patients with type 2 diabetes. However, ADA has no specific recommendations on intermittent fasting for these patients. This article presents a case of a patient who successfully discontinued type 2 diabetes medications by following a low-carbohydrate diet and intermittent fasting.

Summary: This study aims to analyze the benefits of intermittent fasting (IF) combined with a low-carbohydrate diet (LCD) on microvascular and macrovascular outcomes in prediabetic patients. The results show that in the IF and LCD group, there were significant reductions in body weight, waist circumference, body fat percentage, and glycated hemoglobin, as well as a significantly lower incidence of progression to diabetes. Furthermore, the IF and LCD group had a lower incidence of microvascular and macrovascular complications.

Epigenetics of type 2 diabetes mellitus and weight change - a tool for precision medicine?

Charlotte Ling et al.

Summary: This review discusses the role of epigenetic mechanisms in the development of type 2 diabetes mellitus (T2DM) and the regulation of body weight in humans. It highlights the potential of epigenetics in contributing to precision medicine. Studies have shown links between epigenetic modifications and T2DM, as well as non-genetic factors associated with the disease. Genetic factors also influence the epigenome in human tissues. Translational studies have identified blood-based epigenetic markers that could be used for precision medicine in T2DM treatment and risk assessment.

Lifestyle Modifies the Diabetes-Related Metabolic Risk, Conditional on Individual Genetic Differences

Summary: By analyzing data from the UK Biobank, we found a correlation between individual-level lifestyle modifications and changes in diabetes-related metabolic traits. Physical activity was found to modulate the genetic effects on glucose, HbA1c, C-reactive protein, and systolic blood pressure, while alcohol and smoking modulated the effects on BMI, glucose, waist-hip ratio, and diastolic and systolic blood pressure. Additionally, we observed significant phenotypic modulations by lifestyle covariates that were not attributed to genetic effects in the model.
