Induction of autophagy via the PI3K/Akt/mTOR signaling pathway by Pueraria flavonoids improves non-alcoholic fatty liver disease in obese mice

Chunbin Sun et al.

Summary: Non-alcoholic fatty liver disease (NAFLD) is the most common lipid metabolism disorder. Autophagy plays a crucial role in lipid metabolism in NAFLD. Pueraria flavonoids, the main active components of Pueraria lobata, have antioxidant and anti-inflammatory effects. This study demonstrates that Pueraria flavonoids can reduce intracellular lipid accumulation and inflammation levels in NAFLD by stimulating autophagy through the PI3K/Akt/mTOR signaling pathway.

Summary: The American Cancer Society predicts that there will be 1,958,310 new cancer cases and 609,820 cancer deaths in the United States in 2023. While incidence trends for most cancers are favorable, prostate cancer saw a significant increase of 3% annually from 2014 to 2019. However, the overall cancer death rate continues to decline due to advances in treatment, with a 33% reduction since 1991.

Wild pink bayberry free phenolic extract induces mitochondria-dependent apoptosis and G0/G1 cell cycle arrest through p38/MAPK and PI3K/Akt pathway in MDA-MB-231 cancer cells

Wen Xia et al.

Summary: Polyphenol-rich foods have been shown to be effective in preventing cancer due to their powerful antioxidant properties. This study examined the mechanisms by which wild pink bayberry free phenolic extract (WPBFE) inhibits the proliferation and induces apoptosis of MDA-MB-231 breast cancer cells. The results showed that WPBFE down-regulates expression of PCNA, CDK4, cyclin D1, and up-regulates the expression of p21, leading to cell cycle arrest in the G0/G1 phase. WPBFE also induces apoptosis through the mitochondrial death pathway by up-regulating cleaved caspase-3 and enhancing the ratio of Bax/Bcl-2. Furthermore, WPBFE regulates the p38 MAPK and PI3K/Akt pathways.

Summary: Curcumin, isolated from Curcuma longa, has potent tumor-suppressor activity and can decrease the adverse impacts of cisplatin (CP) while enhancing its antitumor activity. Curcumin reduces ROS levels to prevent apoptosis in normal cells, inhibits inflammation, and reduces various toxicities caused by CP. Additionally, curcumin potentiates CP cytotoxicity and suppresses tumor-promoting pathways to prevent drug resistance.

Summary: Phytochemicals, including flavonoids, found in fruits, vegetables, nuts, and legumes have been observed to possess health benefits and can potentially reduce the risk of cancer, heart disease, and neurodegenerative diseases. This review compares the effects and mechanisms of action of several well-characterized and publicized flavonoids, highlighting their potential therapeutic effects. These flavonoids regulate the cell cycle, autophagy, apoptosis, inflammation, and free radicals levels, making them protective of healthy cells but toxic to cancer cells.

Summary: In this study, it was found that prunetinoside (PUG), a plant-based compound, possesses anti-inflammatory effects on mouse macrophage cells. PUG decreased the expression of inflammatory mediators and reduced the activation of phosphorylated signaling pathways. By blocking the NF-κB pathway and activating JNK, PUG could be a potential therapeutic compound for inflammatory-related conditions.

The interplay between apoptosis and cellular senescence: Bcl-2 family proteins as targets for cancer therapy

Alakananda Basu

Summary: The interplay between apoptosis and senescence is crucial in cancer development, and targeting the Bcl-2 family proteins could be a promising strategy for cancer therapy. Therapy-induced senescence (TIS), induced by chemotherapeutic agents, has been controversial in its effect on therapeutic outcome. Clearance of senescent cells and overcoming their pro-survival mechanisms are important challenges in cancer treatment. This review article discusses recent literature on the role of the Bcl-2 family proteins in apoptosis and senescence, and the progress and limitations in targeting them for cancer therapy.

Natural flavonoid luteolin promotes the differentiation of porcine myoblasts through activation of PI3K/Akt/mTOR signaling

Xin Guan et al.

Summary: This study investigated the effects of four representative flavonoids on porcine myoblasts and found that these flavonoids had relatively low cytotoxicity but showed a weak ability to promote cell proliferation. Luteolin, one of the flavonoids, was found to have a positive effect on migration and differentiation of porcine myoblasts. The activation of the PI3K/Akt/mTOR signaling pathway was identified as the mechanism behind luteolin's effects. This study provided biological evidence for the application of luteolin in cultured meat production.
