Surface Engineering of Bioactive Coatings for Improved Stent Hemocompatibility: A Comprehensive Review

Surface modification of biodegradable magnesium alloy with poly (L-lactic acid) and sulfonated hyaluronic acid nanoparticles for cardiovascular application

Summary: Researchers have developed a multifunctional hybrid coating to improve the endothelialization and corrosion resistance of magnesium (Mg) alloy vascular stents. The coating consists of a layer of magnesium fluoride (MgF2) for better corrosion resistance, sulfonated hyaluronic acid (S-HA) nanoparticles for pro-endothelial and anti-inflammatory effects, and poly-L-lactic acid (PLLA) for coating preparation. Blood and cell tests showed that the composite coating had good blood compatibility and promoted endothelial cell growth. This study provides a promising strategy for surface modification of Mg-based cardiovascular stents.

Paclitaxel exerts antiplatelet and antithrombotic activities: Additional benefit from use of paclitaxel-coated balloons and -eluting stents in coronary revascularization and prevention of in-stent restenosis

Summary: This study investigated the antiplatelet activity of paclitaxel in human platelets. The results showed that paclitaxel inhibited platelet aggregation induced by collagen and blocked the downstream signaling molecules of collagen receptor GPVI. It also prevented granule release and GPIIbIIIa activation induced by collagen and attenuated pulmonary thrombosis. Therefore, paclitaxel may have additional benefits when used in drug-coated balloons and drug-eluting stents for coronary revascularization and prevention of in-stent restenosis.

Bioactive Ibuprofen-Loaded PLGA Coatings for Multifunctional Surface Modification of Medical Devices

A rechargeable anti-thrombotic coating for blood-contacting devices

Summary: This study demonstrates a method to regenerate anti-thrombogenic coatings using evolved sortase A to mediate reversible transpeptidation, effectively immobilizing or removing recombinant thrombomodulin and ultra-low molecular weight heparin on a surface. The generation of activated protein C and inhibition of activated factor X were consistent with the molecular composition of the surface, and successful reconstitution of the surface bound coating was shown in a rat model 4 weeks after implantation.

A tailored extracellular matrix (ECM)-Mimetic coating for cardiovascular stents by stepwise assembly of hyaluronic acid and recombinant human type III collagen

Summary: A new recombinant human type III collagen (hCOLIII) without platelet binding sites but with affinity for endothelial cells was explored, together with hyaluronic acid (HA), to form an ECM-mimetic multilayer coating. The coating demonstrated significant thromboprotective properties and guided vascular cell fate by supporting endothelial cell proliferation and inhibiting smooth muscle cell proliferation. Implantation of a polylactic acid (PLA) stent coated with hCOLIII in rabbit abdominal aorta showed enhanced endothelialization, suppressed inflammatory response, inhibition of neointimal hyperplasia, and superior thromboprotection, indicating the potential of hCOLIII-based ECM-mimetic coating as a tailored blood-contacting material for cardiovascular stents.
