Hallmarks of Cancer: New Dimensions

Douglas Hanahan

Summary: The concept of the hallmarks of cancer is a tool for simplifying the complexity of cancer phenotypes and genotypes into basic principles. This article proposes phenotypic plasticity and disrupted differentiation as distinct hallmark capabilities, and discusses nonmutational epigenetic reprogramming and polymorphic microbiomes as enabling characteristics. Additionally, the importance of senescent cells in the tumor microenvironment is highlighted. The integrative concept of the hallmarks of cancer helps in understanding the mechanisms of cancer development and applying that knowledge to cancer medicine.

Fertility sparing surgery in sex-cord stromal tumors: oncological and reproductive outcomes

Alice Bergamini et al.

Summary: Sex cord stromal tumors are rare neoplasms that mainly occur in young women. For patients who want to preserve fertility, unilateral salpingo-oophorectomy is a safe alternative treatment. The spontaneous conception rate is high in granulosa cell tumors, while it is lower in Sertoli-Leydig cell tumors.

Single-cell dissection of cellular components and interactions shaping the tumor immune phenotypes in ovarian cancer

Milena Hornburg et al.

Summary: This study provides a detailed analysis of distinct T cell infiltration patterns in ovarian tumors, revealing the impact on responses to cancer immunotherapies and the underlying key determinants and biology of tumor immune phenotypes. The research highlights potential molecular mechanisms shaping these phenotypes and may inform therapeutic strategies for enhancing clinical benefit from cancer immunotherapies.

Characterizing the tumor microenvironment of metastatic ovarian cancer by single-cell transcriptomics

Susan Olalekan et al.

Summary: Understanding the cellular composition and interactions within the tumor microenvironment is crucial for successful cancer immunotherapies. By performing single-cell RNA sequencing of cells from six ovarian cancer patients, we identified nine major cell types and stratified immune cells into high T cell infiltration and low T cell infiltration groups. The study provides important insights into the immune response in ovarian tumors, highlighting potential anti-tumor responses in the high T cell infiltration group.
