Visible Light-Induced Metal-Free Fluoroalkylations

Summary: Fluoroalkylation is an important synthetic process for modifying molecules with fluoroalkyl groups, which can enhance compound properties and have potential applications in medicine and materials science. The use of visible light-induced, metal-free methods is particularly important due to its environmental friendliness and elimination of potential risks associated with metal-catalyst toxicity. This Account describes our studies on visible light-induced, metal-free fluoroalkylation processes, using organic photocatalysts or EDA complexes. Various pi-systems have been successfully fluoroalkylated under the developed reaction conditions.

A Metal-Free Path to 2-lodo-3-alkyl-1-arylbut-2-en-1-ones and Their Application to the Domino Synthesis of Functionalized 2H-Pyran-2-ones

Summary: We report a metal-free selective synthesis of 2-iodoenones from propargylic alcohols enabled by N-iodosuccinimide. The developed methodology also allows for the synthesis of 3,5,6-trisubstituted 2H-pyran-2-ones through a series of reactions including intermolecular Heck coupling, base-driven Michael addition, and base-mediated double bond isomerization followed by cyclo-condensation.

Nickel-Catalyzed Enantioselective Synthesis of 2,3,4-Trisubstituted 3-Pyrrolines

Summary: The synthesis of highly functionalized chiral 3-pyrrolines is important for the production of natural and synthetic bioactive molecules. Previous methods using allenoates could not synthesize 3,4-disubstituted 3-pyrrolines. This study presents a novel approach to produce 2,3,4-trisubstituted chiral 3-pyrrolines through a highly stereoselective process using a Ni-II/Fc-i-PrPHOX catalytic system.

N-O Bond Activation by Energy Transfer Photocatalysis

Stereoconvergent and -divergent Synthesis of Tetrasubstituted Alkenes by Nickel-Catalyzed Cross-Couplings

Summary: A method to selectively access tetrasubstituted alkenes via nickel-catalyzed Suzuki-Miyaura cross-couplings has been developed, allowing for selective synthesis of each product isomer by judicious choice of ligand and reaction conditions. High-throughput optimization of monophosphine ligands was guided by chemical space analysis, leading to yields up to 94% and diastereomeric ratios up to 99:1 Z/E. Detailed computational and mechanistic studies provided key insights into the isomerization process impacting selectivity in the cross-coupling.

Gas chromatography-mass spectrometry based 18O stable isotope labeling of Krebs cycle intermediates

Summary: A new H-2[O-18] based stable isotope marker was introduced for tracking oxygen exchange rates in different segments of Krebs cycle, increasing selectivity and sensitivity. The method was successfully applied to track the cancer-induced shift in Krebs cycle dynamics in Caco-2 cells, revealing the Warburg effect. The O-18 labeling of Krebs cycle metabolites expands the techniques for monitoring cellular metabolism dynamics.

Direct α-Acylation of Alkenes via N-Heterocyclic Carbene, Sulfinate, and Photoredox Cooperative Triple Catalysis

Summary: N-Heterocyclic carbene (NHC) catalysis is a versatile tool in modern synthetic chemistry, often used in concert with a second catalytic moiety to expand reaction scope. Triple catalysis, merging three different catalytic modes in a single process, has been recently studied, allowing for the synthesis of complex compounds. In a reported study, alpha-C-H acylation of alkenes using NHC, sulfinate, and photoredox cooperative triple catalysis was achieved, highlighting the potential of such synergistic catalytic systems.

Synergistic Effects of Boron and Oxygen Interaction Enabling Nickel-Catalyzed Exogenous Base-Free Stereoselective Arylvinylation of Alkynes through Vinyl Transposition

Summary: This study presents a novel approach to achieve stereoselective synthesis of highly functionalized 1,3-diene products using a stereoselective Ni-catalyzed alkyne vinylation process without the need for exogenous base. Experimental and theoretical studies support the proposed mechanism involving generation of Ni(0) species in situ and subsequent oxidative cyclization with B-assisted C-O bond cleavage. The highly functionalized products have the potential to serve as important building blocks for synthesizing complex organic architectures.
