Nickel-Catalyzed Reductive Coupling of γ-Metalated Ketones with Unactivated Alkyl Bromides

Ning Cui et al.

Summary: A nickel-catalyzed reductive cross-coupling reaction has been developed for the synthesis of aryl alkyl ketones from aryl cyclopropyl ketones and unactivated alkyl bromides. The reaction proceeds via radical cleavage of the alkyl bromide, and provides access to a variety of gamma-alkyl-substituted ketones through ring opening of cyclopropyl ketones (26 examples, 50-90% yield).

Creating Stereocenters within Acyclic Systems by C-C Bond Cleavage of Cyclopropanes

Yair Cohen et al.

Summary: Recent advancements in the stereoselective synthesis of polysubstituted cyclopropanes have enabled chemists to access these strained rings with high diastereomeric and enantiomeric ratios. This development has led to a paradigm shift in the synthesis of stereodefined acyclic molecules through selective carbon-carbon bond cleavage. Application of these concepts has been demonstrated in the total syntheses of various natural products and important molecules, highlighting the power of these strategies in organic synthesis.

A Diverse Library of Chiral Cyclopropane Scaffolds via Chemoenzymatic Assembly and Diversification of Cyclopropyl Ketones

Donggeon Nam et al.

Summary: This study presents a chemoenzymatic strategy for the stereoselective assembly and diversification of cyclopropyl ketones, a versatile class of functionalized cyclopropanes. By utilizing an engineered variant of sperm whale myoglobin, highly diastereo- and enantioselective construction of these molecules is achieved via olefin cyclopropanation in the presence of a diazoketone carbene donor reagent. The resulting enzymatic products can be further transformed to yield a collection of structurally diverse cyclopropane-containing scaffolds in optically pure form, demonstrating the potential of chemoenzymatic synthesis for medicinal chemistry and drug discovery.

Three-Component Coupling of Acyl Fluorides, Silyl Enol Ethers, and Alkynes by P(III)/P(V) Catalysis

Hayato Fujimoto et al.

Summary: The phosphine-catalyzed hydrovinylation reaction involves a three-component coupling of acyl fluorides, silyl enol ethers, and alkynoates. The success of the reaction is attributed to the formal transmetalation between pentacoordinate P(V) species and a silyl enol ether, enabling C-C bond formation between polarity-mismatched sites. This bond formation, not achievable by transition metal catalysis, is achieved through a P(III)/P(V) manifold.

Tunable and Practical Homogeneous Organic Reductants for Cross-Electrophile Coupling

David J. Charboneau et al.

Summary: A series of new tunable homogeneous organic reductants based on a tetraaminoethylene scaffold have been synthesized, which exhibit enhanced air stability and solid state properties at room temperature. These reductants show significantly milder reduction potential than conventional reductants, allowing for controlled alkyl radical generation in Ni-catalyzed XEC reactions. The new reductants are expected to replace current homogeneous reductants due to their stability and lead to the development of novel synthetic methods.

Enantioselective α-Arylation of Ketones via a Novel Cu(I)-Bis(phosphine) Dioxide Catalytic System

Margarita Escudero-Casao et al.

Summary: A novel catalytic system based on copper(I) and chiral bis(phosphine) dioxides has been developed for the arylation of silyl enol ethers, yielding enolizable alpha-arylated ketones with high yields and enantiomeric excess up to 95%. This method is applicable to noncyclic ketones and complements other palladium-catalyzed approaches.
