Synthesis of planar chiral ferrocenes via enantioselective remote C-H activation

Summary: In this study, a modular platform for constructing planar chirality in 1,3-disubstituted ferrocenes/ruthenocenes via an enantioselective relay remote C-H activation strategy was developed. Various 1,3-disubstituted planar chiral metallocenes were prepared with high enantioselectivity (-96-. 99% e.e.). The resulting metallocenes can be easily derivatized for use as ligands and catalysts in asymmetric catalysis and as building blocks for other applications.

Atroposelective remote meta-C-H activation

Summary: In this study, a highly selective remote meta-C-H arylation strategy was reported for the preparation of distally substituted axially chiral scaffolds using a chiral norbornene as a transient mediator. This method provides an efficient route to meta-substituted, axially chiral anilines with broad substrate scope, high stereoinduction, and scalability. The enantioenriched anilines can be readily transformed into diverse chiral compounds, including chiral ligands and catalysts.

Chiral Phosphoric Acid-Palladium(II) Complex Catalyzed Asymmetric Desymmetrization of Biaryl Compounds by C(sp3)-H Activation

Summary: Desymmetrization is a crucial technique for synthesizing chiral compounds, especially chiral biaryls. This study presents an enantioselective synthesis of axially chiral biaryls through desymmetrization using chiral palladium phosphate as the catalyst for C(sp(3))-H activation. Mechanistic investigations reveal that C-H activation plays a crucial role in determining the rate and enantiomeric selectivity. To the best of our knowledge, this is the first report on the asymmetric desymmetrization of axially chiral compounds via C(sp(3))-H activation.

Palladium-Catalyzed C-H Arylation/Arene Dearomatization Domino Reaction: Expeditious Access to Spiro[4,5]fluorenes

Summary: In this study, a palladium-catalyzed C-H arylation/arene dearomatization reaction was performed in a redox-neutral manner. This domino reaction involved in situ-formed Pd(II) species, phenolic-group-directed C-H activation, biaryl cross-coupling, and naphthol dearomatization, leading to the rapid assembly of spiro[4,5]fluorenes with high yield and good chemoselectivity. Interestingly, the malononitrile-derived spirofluorene 6 showed mechanoresponsive luminescence and its potential application in optical memory devices.

Chiral Ligands Based on Binaphthyl Scaffolds for Pd-Catalyzed Enantioselective C-H Activation/Cycloaddition Reactions

Thioether-enabled palladium-catalyzed atroposelective C-H olefination for N-C and C-C axial chirality

Summary: A highly selective C-H olefination reaction was achieved using thioethers as the substrates and a palladium/chiral phosphoric acid catalytic system under ambient air. Axially chiral (hetero)biaryls were successfully synthesized, leading to a broad range of chiral heterocycles with excellent enantioselectivities up to 99% ee. Experimental and computational studies were conducted to understand the mechanism of this reaction and a plausible chiral induction model was proposed based on detailed DFT calculations.

This Review addresses these issues and offers a guide to identify retrosynthetic opportunities to generate C-C bonds by C-H activation processes. Comparing traditional approaches and recent C-H activation methods, this Review demonstrates more efficient retrosynthetic strategies enabled by C-H activation for C-C bond construction.
