Porous microneedle patch with sustained delivery of extracellular vesicles mitigates severe spinal cord injury

Comparison of extruded cell nanovesicles and exosomes in their molecular cargos and regenerative potentials

Summary: Extracellular vesicles (EVs) generated from mesenchymal stem cells (MSCs) play a crucial role in cell-cell communication and tissue regeneration. However, their clinical potential is limited due to low yield. This study compared MSC nanovesicles (NVs) obtained through extrusion with EVs from natural secretion. Proteomics and RNA sequencing data showed that NVs closely resemble MSCs and contain microRNAs associated with cardiac repair, fibrosis repression, and angiogenesis. Intravenous delivery of MSC NVs improved heart repair and cardiac function in a mouse model of myocardial infarction.

Silk Microneedle Patch Capable of On-Demand Multidrug Delivery to the Brain for Glioblastoma Treatment

Summary: A silk fibroin microneedle (SMN) patch designed for glioblastoma (GBM) can bypass the blood-brain barrier and release multiple drugs directly to the tumor site, providing an effective treatment strategy for brain tumors. The biocompatible and biodegradable patch allows for slow drug release over time and can be triggered remotely for rapid drug delivery. In animal models, the patch resulted in decreased tumor volume and increased survival rate, showing potential for clinical treatment of brain tumors.

Immunomodulatory and regenerative effects of the full and fractioned adipose tissue derived stem cells secretome in spinal cord injury

Summary: Adipose tissue derived stem cells (ASCs) secrete molecules that modulate inflammatory response, promote axonal growth, vascular remodeling, and cellular survival. Summary: Functional recovery after traumatic spinal cord injury (SCI) remains a difficult problem to solve due to the limited efficacy of current clinical treatment strategies. In this research, exosomes were engineered through hypoxia stimulation to mesenchymal stem cells and transplanted into the injured spinal cord via encapsulation in a peptide-modified adhesive hydrogel for promoting angiogenesis in SCI therapy. Significant angiogenesis and functional recovery after injury were demonstrated both in vitro and in vivo, indicating the immense potential of hydrogel-encapsulated hypoxia-stimulated exosomes in treating central nervous system trauma and other ischemia diseases.

Local delivery of USC-derived exosomes harboring ANGPTL3 enhances spinal cord functional recovery after injury by promoting angiogenesis
