Conformational heterogeneity of molecules physisorbed on a gold surface at room temperature

Summary: We demonstrate quantitative hyperspectral TERS imaging of single molecules at room temperature, using a freeze-frame approach and a thin Al2O3 capping layer to suppress spectral diffusions and inhibit chemical reactions and contamination. The observed Raman peak variation reveals conformational heterogeneity at the single-molecule level. This work extends the scope of single-molecule vibrational spectroscopy studies.

Proteostasis of Islet Amyloid Polypeptide: A Molecular Perspective of Risk Factors and Protective Strategies for Type II Diabetes

Summary: The possible link between hIAPP accumulation and beta-cell death in diabetic patients has sparked a lot of research on amyloid structures and aggregation pathways. Recent studies have highlighted the importance of early oligomeric intermediates and their interactions with lipid membranes, pH, insulin, and zinc in the mechanism of hIAPP aggregation. Challenges in characterizing the aggregation process have led to the development of various biophysical and chemical approaches, with studies suggesting that restoring hIAPP proteostasis could be a promising strategy for therapy.

Single Molecule Surface Enhanced Raman Scattering in a Single Gold Nanoparticle-Driven Thermoplasmonic Tweezer

Tracking interfacial single-molecule pH and binding dynamics via vibrational spectroscopy

High-speed field-enhanced spectroscopy allows for monitoring chemical switching of a single carboxylic group between three discrete states, with theoretical calculations identifying reversible proton transfer dynamics and switching between molecule-metal coordination states. Recent advances in plasmonic nanocavities for single-molecule spectroscopy

The extreme sensitivity is made possible by highly confined local field intensity enhancement, which is dependent on the geometry of plasmonic nanocavities.Suitably designed structures that provide engineered local optical fields lead to enhanced optical sensing based on phenomena like surface enhanced Raman scattering, fluorescence, and Forster resonance energy transfer.
