Emerging roles for RNA-binding proteins in T lymphocytes

Silvia Monticelli

Summary: RNA-binding proteins play a crucial role in regulating gene expression in the immune system, especially in T lymphocytes. Many newly discovered RNA-binding proteins still have unknown functions, regulation, and roles in gene regulatory networks involved in immune responses. Studying the emerging role of unconventional RNA-binding proteins and distinguishing functional RNA-binding protein:mRNA interactions from those with limited impact are essential for fully understanding the complexities of RNA-binding protein-mediated regulation in the immune system.

The function of Wtap in N6-adenosine methylation of mRNAs controls T cell receptor signaling and survival of T cells

Taku Ito-Kureha et al.

Summary: The m(6)A modification plays a crucial role in the development, activation and survival of T cells. The Wtap protein is essential for m(6)A methyltransferase complex function and is crucial in TCR signaling. m(6)A modification destabilizes Orai1 and Ripk1 mRNAs, impacting the survival of T cells.

Wilms' tumor 1-associating protein complex regulates alternative splicing and polyadenylation at potential G-quadruplex-forming splice site sequences

Keiko Horiuchi et al.

Summary: The WTAP complex regulates alternative splicing by promoting exon skipping and intron retention, especially in AS events involving short introns/exons with higher GC content and weaker polypyrimidine-tract and branch points. Additionally, the interaction between WTAP and CBLL1 facilitates the recruitment of the 30-end processing complex, indicating an inhibitory mechanism in GC-rich sequences for AS and alternative polyadenylation.

Small-molecule inhibition of METTL3 as a strategy against myeloid leukaemia

Eliza Yankova et al.

Summary: The study identified a highly potent and selective catalytic inhibitor of METTL3, STM2457, which shows potential therapeutic value in treating AML. Treatment with this inhibitor leads to reduced AML growth, increased differentiation and apoptosis, and decreased m(6)A levels on leukemogenic mRNAs. In vivo pharmacological inhibition of METTL3 results in impaired engraftment and prolonged survival in mouse models of AML.

RNA-binding proteins and RNA methylation in myeloid cells*

Marian Bataclan et al.

Summary: RNA-binding proteins (RBPs) play crucial roles in regulating mRNA lifecycle, immune responses, and pathogen sensing, with dysregulated activity potentially leading to diseases. The expression and function of RBPs are tightly controlled by a complex network of regulatory mechanisms, including cross-regulation of different RBPs.

Deep and accurate detection of m6A RNA modifications using miCLIP2 and m6Aboost machine learning

Nadine Koertel et al.

Summary: The combination of miCLIP2 with machine learning significantly improves m(6)A detection, allowing for high-complexity libraries with less input material. The development of the m6Aboost model enables the prediction of genuine m(6)A sites without the need for filtering for specific sequences. The methodology provides a rich resource for future analysis of m(6)A sites in different cell lines.
