Lipotoxicity-induced STING1 activation stimulates MTORC1 and restricts hepatic lipophagy

Kunpeng Liu et al.

Summary: The study identified a novel regulatory loop between STING1 and MTORC1, explaining how hepatic inflammation regulates lipid accumulation. The findings may facilitate the development of new strategies for clinical treatment of hepatic steatosis.

Summary: The liver plays a crucial role in immune defense during acute systemic infections. This study identified a subpopulation of proinflammatory hepatocytes that have key immunoregulatory roles in endotoxemia. These findings provide a conceptual framework for discovering rational therapeutic targets in acute infection.

Replication stress triggered by nucleotide pool imbalance drives DNA damage and cGAS-STING pathway activation in NAFLD

Romain Donne et al.

Summary: This study investigated the mechanisms of DNA damage response during NAFLD and found that nucleotide pool imbalance is the key driver of replication stress (RS), and DNA damage also activates the cGAS/STING pathway, promoting disease progression.

Hepatic cytochrome P450 8B1 and cholic acid potentiate intestinal epithelial injury in colitis by suppressing intestinal stem cell renewal

Li Chen et al.

Summary: This study reveals that disrupted bile acid (BA) homeostasis, specifically increased cholic acid (CA) levels and induced cytochrome P450 8B1 (CYP8B1) synthesis, plays a significant role in the pathogenesis of colitis. CA inhibits peroxisome proliferator-activated receptor alpha (PPAR alpha), resulting in impaired fatty acid oxidation (FAO) and intestinal stem cell (ISC) renewal, leading to more severe colitis. Activation of the farnesoid X receptor (FXR) suppresses CYP8B1 expression and ameliorates colitis. These findings suggest that targeting the hepatic CYP8B1-CA axis may be a beneficial strategy for treating inflammatory bowel disease (IBD).

Targeted therapeutics and novel signaling pathways in non-alcohol-associated fatty liver/steatohepatitis (NAFL/NASH)

Xiaohan Xu et al.

Summary: Non-alcohol-associated fatty liver/steatohepatitis (NAFL/NASH) has become the leading cause of liver disease worldwide, with a higher risk of developing cirrhosis and hepatocellular carcinoma. Lifestyle interventions are currently the most effective strategies for preventing and controlling NAFL without fibrosis. Promising progress has been made in understanding the pathogenesis and identifying therapeutic targets. The disruption of the gut-liver axis and microbe-derived metabolites play a vital role in driving NAFL/NASH. Targeting gut microbiota or extracellular vesicles (EVs) may serve as new strategies for NAFL/NASH treatment.

RNA-Binding RING E3-Ligase DZIP3/hRUL138 Stabilizes Cyclin D1 to Drive Cell-Cycle and Cancer Progression

Srinivasa P. Kolapalli et al.

Summary: This study demonstrates, for the first time, that DZIP3 uses a unique two-pronged mechanism to stabilize Cyclin D1 and drive cell-cycle progression and cancer progression. DZIP3 controls Cyclin D1 mRNA and protein stability in a cell-cycle phase-dependent manner, making it a novel driver of cell-cycle and cancer progression.

Mechanisms and disease consequences of nonalcoholic fatty liver disease

Rohit Loomba et al.

Summary: NAFLD and NASH are leading chronic liver diseases worldwide, causing progressive liver injury that can lead to cirrhosis and hepatocellular carcinoma. These diseases are associated with hepatic glucose and lipid metabolism, and are influenced by genetic, epigenetic, and environmental factors.

Non-alcoholic fatty liver disease

Elizabeth E. Powell et al.

Summary: Non-alcoholic fatty liver disease (NAFLD) is a common global health issue with significant associations with metabolic syndrome and type 2 diabetes, leading to serious complications like cirrhosis and hepatocellular carcinoma. While cardiovascular disease and extrahepatic malignancy are the main causes of death in NAFLD patients, advanced liver fibrosis is a key prognostic marker for liver-related outcomes and overall mortality.

Tripartite motif 16 ameliorates nonalcoholic steatohepatitis by promoting the degradation of phospho-TAK1

Lin Wang et al.

Summary: The study identified TRIM16 as a promising therapeutic target for NASH by demonstrating its ability to alleviate lipid accumulation and inflammation in the liver, as well as suppress the activation of the MAPK signaling pathway. This suggests that TRIM16 could be a potential candidate for treating NASH-related liver disorders and hepatocellular carcinoma.
