Evolution of the Neocortex Through RNA-Binding Proteins and Post-transcriptional Regulation

Iva Salamon et al.

Summary: The evolution and development of the human neocortex rely on the abundance, diversity, and temporal molecular regulation of progenitor cells. RNA-binding proteins play important roles in different stages of early neurogenesis, particularly in the cell fate transition of transcriptionally primed cortical progenitors.

Synaptic Vesicle Glycoprotein 2A: Features and Functions

Rachele Rossi et al.

Summary: SV2A, a transmembrane protein of synaptic vesicles, serves as a potential marker of synaptic density with stronger associations to GABAergic synapses. Despite being ubiquitously expressed, the biological function and interaction between SV2A and its tracers remain unclear. Further research is required for a better understanding of the protein's involvement in neurotransmission and its implications in neuroimaging studies.

Human brain evolution: Emerging roles for regulatory DNA and RNA

Jing Liu et al.

Summary: Humans differ from other primates in terms of neuroanatomy and cognitive functions, with human-specific features being particularly prominent in the cerebral cortex due to an expansion in size and unique cellular composition and circuitry. It is postulated that human-specific gene expression may explain neocortical anatomical differences across evolution, particularly in relation to noncoding regulatory loci linked to human traits. The review also highlights emerging noncoding elements in human cortical evolution and their associations with neurodevelopmental diseases.

Parallel in vivo analysis of large-effect autism genes implicates cortical neurogenesis and estrogen in risk and resilience

Helen Rankin Willsey et al.

Summary: Gene Ontology analyses have repeatedly identified synaptic function and transcriptional regulation as key points of convergence in autism spectrum disorders (ASD) risk genes. Leveraging Xenopus tropicalis, a study of ten genes associated with ASD found that mutations led to an increase in neural progenitor cells, implicating cortical neurons in ASD vulnerability and expanding the range of convergent functions to include neurogenesis. Systematic chemical screening identified estrogen, through Sonic hedgehog signaling, as a resilience factor that may mitigate a range of ASD genetic risks in Xenopus and human brain models.

Decoding mixed messages in the developing cortex: translational regulation of neural progenitor fate

Mariah L. Hoye et al.

Summary: Regulation of stem cell fate decisions is crucial for development, homeostasis, and organismal fitness. Transcriptional priming of neural progenitors and dynamic post-transcriptional regulation, particularly in radial glial progenitors, play a vital role in defining cell fates and are implicated in neurodevelopmental disease. Post-transcriptional mechanisms, including subcellular mRNA localization, RNA decay, and translation, are highlighted in governing cortical development and translational control of neuronal fates.
